雌激素通过ER-α调控CYP2C8在鸡肝中的表达

【摘要】:本研究旨在探讨鸡CYP2C8基因的表达调控规律。采集30周龄卢氏绿壳蛋鸡心、肝、脾、肺、肾、胸肌、腹脂、肌胃、腺胃、卵巢、胰、十二指肠、空肠、回肠14种组织以及从1日龄~30周龄不同发育时期的肝组织,用荧光实时定量PCR分析CYP2C8基因的时空表达规律;用雌激素及其受体拮抗剂对活体和体外培养的鸡胚肝原代细胞进行处理,用荧光实时定量PCR分析CYP2C8基因的表达调控机制。结果表明,鸡CYP2C8基因在各组织中表现出广泛表达的特性,但在脂类代谢旺盛的器官如肝、肾、小肠(十二指肠、空肠、回肠)等器官中表达较高,且在肝中的表达水平随着性成熟而显著升高(P0.01);雌激素处理可显著提高鸡肝组织和体外培养的鸡胚胎肝原代细胞中CYP2C8的表达水平(P0.05),但雌激素受体ER-α拮抗剂MPP可完全抑制雌激素的作用(P0.05)。综上表明,鸡CYP2C8基因在不同组织中广泛表达,而以肝中的表达水平最高,且肝中CYP2C8基因的表达是受雌激素并通过ER-α调控。
[Abstract]:The purpose of this study was to investigate the regulation of chicken CYP2C8 gene expression. The heart, liver, spleen, lung, kidney, pectoral muscle, abdominal fat, muscle stomach, glandular stomach, ovary, pancreas, duodenum, jejunum, ileum and liver tissues were collected from 30 week old Luhs green shell laying hens at 30 weeks of age. The temporal and spatial expression of CYP2C8 gene was analyzed by real-time quantitative PCR, estrogen and its receptor antagonists were used to treat the primary cells of chicken embryo liver in vitro and in vivo, and the regulation mechanism of CYP2C8 gene expression was analyzed by real-time quantitative PCR. The results showed that chicken CYP2C8 gene was widely expressed in various tissues, but it was highly expressed in organs with high lipid metabolism such as liver, kidney, small intestine (duodenum, jejunum, ileum) and so on. The expression level of CYP2C8 in liver was significantly increased with sexual maturation, and estrogen treatment could significantly increase the expression of CYP2C8 in chicken liver tissue and in vitro cultured chicken embryonic liver primary cells. However, estrogen receptor ER- 伪 antagonist MPP could completely increase the expression level of P0.05, and estrogen receptor ER- 伪 antagonist MPP could completely increase the expression level of P0.05G in chicken liver tissue and embryonic liver cells in vitro. Inhibitory effect of estrogen on P0.05. In conclusion, chicken CYP2C8 gene was widely expressed in different tissues, but the highest expression level was in liver, and the expression of CYP2C8 gene in liver was regulated by estrogen and ER- 伪.
