西门塔尔杂种牛生长速度相关基因与生长性状关联性的研究

【摘要】:本试验选取西门塔尔杂种牛243头为研究对象,选择5个与生长速度相关基因,根据GenBank公布的牛的序列为模板,共设计了14对引物。利用PCR-SSCP技术测序分析了5个基因部分序列多态性,结果有7对引物(GH-G2、GH-G3、UCP2-U1、UCP2-U2、UCP3-U3、UCP3-U4、NEB-N1)扩增片段存在多态性,并分析了这4个基因的7个片段多态性与生长性状之间的相关性;利用分子生物学软件对基因变异位点进行多态信息分析。研究结果如下:1.西门塔尔杂种牛GH-G2、UCP3-U4、NEB-N1位点PIC值分别为0.1376、0.1281、0.2064,处于低度多态(PIC0.25), GH-G3、UCP2-U1、UCP2-U2、UCP3-U3位点PIC值分别为0.3576、0.4056、0.3491、0.3727,处于中度多态(0.25PIC0.5)。2.GH-G2片段存在204CT、420GA两处突变;GH-G3片段存在3699GC、4000GT、4001AG三处突变,其中两处分别引起亮氨酸变成缬氨酸,缬氨酸变成异亮氨酸;UCP2-U1片段存在632CT、655GA、689CA三处突变,UCP2-U2片段处检测到643AC的突变,均属于同义突变;UCP3-U3片段5465AG,引起丙氨酸被苏氨酸替代;UCP3-U4检测发现四处突变分别为7953GC、7921GA、 8018 CT、8162CT; NEB-N1片段20166AG的突变。3.NEB-N1位点适合用于西门塔尔杂种牛增重速度、体高、胸围分子标记辅助选择,BB型为优势基因型,尤其后期增重效果、胸围优势非常明显;UCP3-U3位点对生长后期增重速度影响显著,可以作为增重分子标记辅助选择;UCP3-U4位点适合用于西门塔尔杂种牛体斜长分子标记辅助选择,AJ3型为优势基因型。
[Abstract]:In this experiment, 243 Simmental hybrid cattle were selected as research objects, 5 genes related to growth rate were selected, and 14 pairs of primers were designed according to the sequence of cattle published by GenBank. The partial polymorphisms of five genes were sequenced by PCR-SSCP technique. The results showed that there were 7 pairs of primers GH-G2HG3UCP2-U1UCP2-U2UCP3-U3C3-UCP3-UCP3-U4NEB-N1). The relationship between the polymorphism of seven fragments of the four genes and the growth traits was analyzed. The polymorphic information of gene mutation sites was analyzed by molecular biology software. The results are as follows: 1. The PIC values of the GH-G2UCP3-U4NB-N1 locus of Simmental hybrid cattle were 0.1376-0.1281nb 0.2064, respectively, and the PIC values of GH-G3UCP2-UCP2-UCP2-UCP2-UCP3-UCP3-U3 were 0.35760.40560.3491N 0.3727, respectively. The mutation GH-G3 at 204CT420GA was 0.35760.40560.3491N 0.3727, and the mutation of GH-G3 at 204CT420GA was 3699GCf4000GT4001AG 3, respectively. The mutation of 643AC was detected at the UCP2-U1 fragment with three mutations of 632CT655GAA689CA and the fragment of UCP2-U2 was changed from valine to isoleucine. All of them belong to the synonymous mutation of UCP3-U3 fragment (5465AGG), which caused alanine to be substituted by threonine for UCP3-U4. The results showed that the four mutations were 7953GCf7921GA and 8018 CT-8162CT.The mutation of NEB-N1 fragment 20166AG. 3. NEB-N1 locus was suitable for Simmental hybrid cattle to gain weight and gain body height. The breast circumference molecular marker-assisted selection of BB genotype was the dominant genotype, especially in the later stage of weight gain. The breast circumference dominance was very obvious. The UCP3-U3 site had a significant effect on the weight gain rate in the late growth period. UCP3-U4 locus can be used as weight gain molecular marker assisted selection of AJ3 genotype in Simmental hybrid cattle.
