猪浓缩饲料蛋白质氧化及不同抗氧化剂抗氧化作用的研究

【摘要】:猪浓缩饲料应用较为普遍,其主要组成部分是蛋白质饲料,含蛋白质较高,通常贮存时间较长,在储存过程中蛋白质易受各种因素影响而发生氧化,从而影响其应用效果。本研究采集饲料企业实际生产的猪浓缩饲料进行蛋白质氧化状况分析,并研究蛋白质发生氧化的浓缩饲料对仔猪的影响;同时研究不同类型抗氧化剂对乳猪浓缩饲料蛋白质氧化的影响,为保证浓缩饲料的品质提供参考依据。论文包括以下3个试验:试验一调研分析部分饲料企业猪浓缩饲料蛋白质氧化状况。选择江苏省有代表性的饲料企业进行浓缩饲料样品采集,根据生产时间、储藏时间等条件,分为不同的批次,将采集的饲料样用四分法缩减至200g左右并粉碎过60目,进行蛋白质氧化指标(蛋白质羰基、琉基、游离氨基含量)测定。结果表明,随着储存时间的延长,浓缩饲料中的蛋白质羰基含量逐渐升高(P0.05),蛋白质游离巯基、总巯基及游离氨基含量逐渐降低(P0.05),提示蛋白质的氧化程度随着储存时间的延长而升高。试验二研究常规条件储存浓缩饲料对仔猪生长性能和血清生理生化指标的影响。选用健康、体重相近的35日龄断奶仔猪64头,随机分为2组,每组4重复,每个重复8头猪,日粮处理如下:(1)日粮所用浓缩饲料为4℃低温储存45 d;(2)日粮所用浓缩饲料为常规条件储存45 d。结果表明,与低温储存组相比,常规条件储存组仔猪平均日增重(ADG)和平均日采食量(ADIF)显著降低(P0.05),仔猪血清谷丙转氨酶(ALT)活性及白蛋白(ALB)含量显著降低(P0.05),血清尿素氮(UN)含量显著升高(P0.05)。试验三研究不同类型抗氧化剂对浓缩饲料中蛋白质氧化的影响。选用乳猪浓缩饲料为试验材料,处理如下:(1)AO1组不添加任何抗氧化剂;(2)AO2组添加固体乙氧基喹淋;(3)AO3组添加液体乙氧基喹淋;(4)AO4组添加固体和液体乙氧基喹啉。添加剂量均为150g/t乙氧基喹淋。将各处理饲料样品分别置于室温25 ℃和模拟夏季高温35 ℃培养箱内储存不同时间(0d,10 d,20 d,30 d和60 d)。结果表明,随着储存时间的延长各组浓缩饲料蛋白质羰基含量显著升高(P0.05),巯基含量显著降低(P0.05),游离氨基含量没有显著变化(P0.05),NSI及蛋白质体外消化率显著降低(P0.05);乙氧基喹淋具备一定的抗氧化作用,储存时间在20 d以前4组之间蛋白质氧化程度没有显著差异,储存30 d以后,液体乙氧基喹淋的抗氧化效果显著优于固体乙氧基喹啉(P0.05),固、液体乙氧基喹啉各半使用的抗氧化效果不稳定。
[Abstract]:Porcine concentrated feed is widely used and its main component is protein feed, which contains high protein and usually has a long storage time. During storage, protein is easily oxidized by various factors, thus affecting its application effect. In this study, the protein oxidation status of pig concentrated feed produced by feed enterprises was analyzed, and the effect of protein oxidized concentrated feed on piglets was studied. At the same time, the effects of different antioxidants on protein oxidation in concentrated feed of suckling pigs were studied in order to guarantee the quality of concentrated feed. The thesis includes the following three experiments: experiment 1: investigation and analysis of protein oxidation status of pig concentrate feed in some feed enterprises. The representative feed enterprises in Jiangsu Province were selected to collect concentrated feed samples. According to the production time, storage time and other conditions, the samples were divided into different batches, and the collected feed samples were reduced to about 200g by the four-part method and crushed over 60 mesh. Protein oxidation index (protein carbonyl, Ryukyl, free amino content) was determined. The results showed that the content of protein carbonyl in concentrated feed increased gradually with the prolongation of storage time, and the protein free sulfhydryl group increased gradually. The content of total sulfhydryl group and free amino group decreased gradually, suggesting that the oxidation degree of protein increased with the prolongation of storage time. The second experiment was to study the effects of conventional storage of concentrated feed on growth performance and serum physiological and biochemical indexes of piglets. Sixty-four weaned piglets of 35 days of similar body weight were randomly divided into 2 groups with 4 replicates in each group and 8 piglets in each replica.
