鸭OIH、FSHβ基因多态性及其与产蛋性状的关联研究

【摘要】:本研究以卵抑制剂(OIH)基因和促卵泡激素(FSHβ)基因为鸭产蛋性状相关候选基因,先克隆获得了两基因部分序列,并采用DNA池技术检测了两基因的DNA多态性,两个基因各选择两个SNP位点做PCR-RFLP,以连城白鸭与白改鸭杂交第3代群体为材料,检测各个体的基因型并研究性状的关联,主要研究结果如下:1.克隆获得了鸭卵抑制剂(OIH)基因Exon10-Exon12和Intorn10-Intorn12的部分序列,采用DNA池加测序的方法对鸭卵抑制剂(OIH)基因编码区及其侧翼序列进行SNP扫描,共检测到4个新的SNP位点,其中位点c.316AG和c.1287 AC分别导致了限制性内切酶Acc I和Alu I酶切位点多态性的改变。在连城白鸭与白改鸭杂交第3世代群体中,c.316AG位点等位基因G和A频率分别为0.33和0.67,c.1287 AC位点等位基因C和A频率分别为0.91和0.09;关联分析结果显示,位点c.316AG与群体的420天产蛋量(P=0.05),蛋壳强度(P0.05)存在显著相关,与390天产蛋量(P=0.08)相关程度接近显著水平;位点c.1287 AC与连城白鸭与白改鸭杂交群体的270天产蛋量(P0.05),300天产蛋量(P0.05),330天产蛋量(P0.01),360天产蛋量(P0.01),390天产蛋量(P0.01),420天产蛋量(P0.01)存在显著相关或极显著相关。2.扩增获得了鸭促卵泡激素(FSHβ)基因第三外显子部分编码区序列;在鸭FSHβ基因序列中共检测到7个新的SNP位点,其中位点c.82CG和c.308TG分别导致Alu I和Ban II酶切位点的改变;在连城白鸭与白改鸭杂交第3世代群体中,c.82CG位点等位基因C和G频率分别为0.24和0.76,c.308TG位点等位基因T和G分别为0.64和0.36;性状关联分析结果表明,位点c.82CG与连城白鸭与白改鸭杂交群体的开产蛋重(P0.01)、蛋白高度(P0.05)存在显著或极显著相关;位点c.308TG与连城白鸭与白改鸭杂交群体的120天产蛋量(P0.01)存在极显著相关,与开产日龄(P=0.06)相关性接近显著水平。3.利用PHASE软件对以上两基因突变位点进行了单倍型重构,在连城白鸭与白改鸭杂交第3世代群体中,性状关联分析结果表明,OIH基因重构二倍体型与连城白鸭与白改鸭杂交群体的后期产蛋量(EP330d,EP360d,EP390d,EP420d)存在显著相关(P0.05),FSHβ基因重构二倍体型与连城白鸭与白改鸭杂交群体的早期产蛋量(EP120d,EP180d,EP210d,EP240d)存在显著(P0.05)或者极显著(P0.01)相关。
[Abstract]:In this study, OIH gene and follicle stimulating hormone (FSH 尾) gene were used as candidate genes for duck egg-laying traits. The partial sequences of the two genes were cloned, and the DNA polymorphisms of the two genes were detected by DNA pool technique. Two SNP loci of each gene were selected as PCR-RFLP.The third generation population of Liancheng White Duck and White modified Duck were used as materials to detect the genotypes of each individual and study the correlation of traits. The main results were as follows: 1. The partial sequences of duck egg inhibitor (OIH) gene Exon10-Exon12 and Intorn10-Intorn12 were cloned. The coding region and flanking sequence of duck egg inhibitor (OIH) gene were scanned by DNA pool plus sequencing, and four new SNP sites were detected. Among them, locus c.316AG and c.1287AC resulted in the polymorphism of restriction endonuclease Acc I and Alu I, respectively. In the third generation population of Liancheng White Duck and White modified Duck, the alleles G and A at allele C and A were 0.33 and 0.67 c. 1287, respectively, and the correlation analysis showed that the allele C and A were 0.91 and 0.09, respectively. Locus c.316AG was significantly correlated with egg production (P0. 05) and eggshell strength (P0.05), and was close to significant level (P0. 08). There was significant correlation or extremely significant correlation between the number of eggs laid in 270 days (P0.05) and the number of eggs laid in 330 days (P0.01), 360 days (P0.01), 390 days (P0.01) and 420 days (P0.01) of Liancheng White Duck (P0.01) and Liancheng White Duck (P0.01). The partial coding region of the third exon of the duck follicle stimulating hormone (FSH 尾) gene was obtained, and seven new SNPs were detected in the FSH 尾 gene sequence, in which c. 82CG and c. 308TG led to the change of Alu I and Ban II endonuclease sites, respectively. In the third generation population of Liancheng White Duck and White modified Duck, the alleles C and G at allele C and G were 0.24 and 0.76 and 0.64 and 0.36, respectively, and the results of trait correlation analysis showed that the allele C and G were 0.64 and 0.36, respectively. Locus c.82CG was significantly correlated with egg weight (P0.01) and protein height (P0.05) of Liancheng white duck crossing population, and locus c.308TG was significantly correlated with 120-day egg production (P0.01) of Liancheng white duck cross population. The correlation with the age of birth (P0. 06) was close to the significant level of 3. 3. Haplotype reconstruction of the mutation sites of the two genes was carried out by phase software in the third generation population of Liancheng White Duck and White modified Duck. The results of trait association analysis showed that there was a significant correlation between the late egg production (EP330d / EP360d / EP390d/ EP420d) between Liancheng White Duck and Liancheng White Duck (P0.05) and Liancheng White Duck (Licheng White Duck) population by FSH 尾 gene reconstruction diploid type and Liancheng white duck cross population (EP330d / EP360d / EP390d / EP420d). Egg production (EP120 d, EP180 d, EP210 d, EP240 d) was significantly (P0.05) or extremely significant (P0.01).
