犬弓首蛔虫线粒体cox1基因测序鉴定及生物学特性研究

【摘要】:犬弓首蛔虫属于蛔目(Ascarididea)弓首科(Toxocaridae)弓首属(Toxocara),寄生于犬和其它动物(包括人)的小肠和胃内的一种线虫。本研究为了了解犬弓首蛔虫虫卵在体外发育情况、鉴别其感染性虫卵,掌握幼虫在小鸡体内移行情况,而做了一系列的相关研究,为犬弓首蛔虫病的诊断和防治提供了大量科学依据。本研究的第一部分对7株犬蛔虫cox1基因进行测序与序列分析。结果显示7株犬蛔虫cox1基因序列长度为440bp。线粒体cox1基因的种内相似性为96.7%~100%,种间相似性为86.4%-90.8%。本研究的第二部分设在5种温度下,采用十种试剂培养方法来体外培养犬弓首蛔虫虫卵,以及对犬弓首蛔虫虫卵在自然生活环境中进行体外模拟培养试验,结果显示,在15℃、25℃、35℃条件下,各体外试剂培养方法组和各模拟试验组的犬弓首蛔虫虫卵均有发育,在5℃、45℃条件下各组均未发育。十种体外试剂培养方法中,以25℃1molH2SO4培养犬弓首蛔虫虫卵孵化最快。模拟实验中,以25℃清水中的犬弓首蛔虫虫卵孵化最快。本研究的第三部分为犬弓首蛔虫感染性虫卵的鉴定和犬弓首蛔虫幼虫在小鸡体内移行试验。结果显示含两圈幼虫期(6字形会动)虫卵和含两圈幼虫期(8字形会动)虫卵为感染性虫卵,其幼胚长度均为238-270μm。在小鸡感染幼虫12小时肝脏最早分离出幼虫,肺脏分离出幼虫最早时间为感染后24h。本研究为国内首次对犬弓首蛔虫虫卵在自然环境进行模拟体外培养试验,以及对犬弓首蛔虫感染性虫卵进行的鉴别,在国内首次犬弓首蛔虫幼虫在小鸡体内移行试验。本研究为犬弓首蛔虫的分子生物学、生物学特性、临床试验以及公共卫生学等诸多研究提供了依据。
[Abstract]:Ascaris canis belongs to the genus Toxocara of the (Ascarididea) family Toxocaridae, a nematode parasitic on the small intestine and stomach of dogs and other animals (including human beings). In order to understand the development of Ascaris canis eggs in vitro, identify their infective eggs, and master the migration of larvae in chickens, a series of related studies were carried out in this study. It provides a great deal of scientific basis for the diagnosis and prevention of Ascaris canis disease. In the first part of this study, 7 cox1 genes of Ascaris canis were sequenced and sequenced. The results showed that the length of cox1 gene of 7 strains of Ascaris canis was 440 BP. The intraspecific similarity of mitochondrial cox1 gene was 96.7 and the interspecific similarity was 86.4-90.8.
