【摘要】:谷胱甘肽过氧化物酶-5 (Glutathione peroxidase type-5)在动物附睾中特异性表达,并对精子的成熟起着至关重要的作用。本文研究了GPX5在不同年龄的绵羊附睾中的表达情况。研究结果如下:1.通过实时荧光定量PCR,检测GPX5基因在不同年龄绵羊附睾,睾丸和输精管的相对表达量。结果表明GPX5基因在幼龄(2-3月龄)羔羊附睾、睾丸和输精管中几乎不表达;而在青年(6-12月龄)绵羊附睾头,GPX5基因的表达量极显著高于附睾体和附睾尾(p0.01),在输精管和睾丸几乎不表达;在成年(2-3岁)绵羊附睾头显著高于附睾体和附睾尾(p0.05),但显著低于青年绵羊附睾头的表达量。GPX5在不同年龄绵羊的睾丸和输精管的表达量没有显著差异。2.通过western blot的方法检测不同年龄绵羊的附睾和睾丸中GPX5蛋白的表达。结果显示在羔羊附睾和睾丸未检测到GPX5蛋白;在青年绵羊附睾头高表达,附睾体和附睾尾中低表达,睾丸中不表达;在成年绵羊附睾头少量表达,附睾体,附睾尾和睾丸几乎都不表达。3.利用免疫组化的方法,对GPX5蛋白在附睾上进行定位。经过免疫组化分析,GPX5蛋白主要在附睾头上皮细胞质以及静纤毛表达,附睾体和附睾尾中的GPX5蛋白主要集中在附睾管腔中,上皮少量表达。并且附睾尾的GPX5蛋白主要在静纤毛表达。GPX5蛋白从附睾上皮分泌到附睾腔中,随着精子在附睾中的运输与精子结合。总之,GPX5在不同年龄的绵羊附睾中的表达存在差异。GPX5主要在青年绵羊和,成年绵羊的附睾头上皮表达,GPX5蛋白分泌到附睾管腔中与精子结合,保护精子在附睾运输过程中不受活性氧(ROS)的攻击,保证精子功能的正常发挥。
[Abstract]:Glutathione peroxidase -5 (Glutathione peroxidase type-5) is specifically expressed in animal epididymis and plays a vital role in the maturation of sperm. This paper studies the expression of GPX5 in the epididymis of sheep at different ages. The results are as follows: 1. by real-time quantitative fluorescence quantitative PCR, the GPX5 gene was detected at different ages.
