一次性腹腔注射Diquat诱导构建肉仔鸡慢性氧化应激模型

【摘要】:通过对肉仔鸡一次性腹腔注射Diquat,研究试验期间肉仔鸡的生长性能、部分血液生化指标和抗氧化指标的变化,建立肉仔鸡慢性氧化应激模型。选取300只20日龄的健康科宝肉仔鸡,随机分为2组,每组15个重复,每重复10只鸡,公母各半。试验期共21d。试验第1天,处理组按照体质量以8mg/kg一次性腹腔注射Diquat,对照组注射相应剂量的无菌生理盐水。所有鸡自由采食和饮水,在试验第1、12、21天,空腹12h,然后前腔静脉采血分离血清,测定血清中的生化指标和抗氧化指标。结果显示,Diquat处理组的平均日增量和平均日采食量显著低于对照组,料体质量比有增高的趋势;血清中ALT、AST、GSH-Px、SOD和COR活性显著高于对照组,MDA浓度显著低于对照组。表明,按照8mg/kg体质量一次性腹腔注射Diquat溶液可以诱导肉仔鸡产生氧化应激,并且应激效应可持续21d,血清中ALT、AST、GSH-Px、COR、SOD和MDA的含量变化可以作为反映氧化应激的敏感指标。
[Abstract]:The chronic oxidative stress model of broilers was established by intraperitoneal injection of Diquat in a single dose to study the changes of growth performance, some biochemical indexes and antioxidant indexes of broilers during the experiment. Three hundred 20-day-old healthy Kebao broilers were randomly divided into two groups, 15 replicates in each group, 10 in each replica, half in male and half in female. The trial period was 21 days. On the first day of the experiment, Diquat was injected intraperitoneally with 8mg/kg according to body mass in the treatment group, and the control group was injected with aseptic saline at the corresponding dose. All chickens were fed and drank freely. The serum samples were collected from the anterior vena cava for 12 hours on the 21st day and 12 hours before the first vena cava. The biochemical and antioxidant indexes in the serum were determined. The results showed that the average daily increment and average daily feed intake of Diquat treatment group were significantly lower than those of the control group, and the ratio of feed to body weight was increasing, and the activities of SOD and cor in serum ALT ASTX GSH-PxN were significantly higher than those in the control group, which were significantly lower than those in the control group. The results showed that the oxidative stress of broilers could be induced by single intraperitoneal injection of Diquat solution according to the body weight of 8mg/kg, and the stress effect could be sustained for 21 days. The changes of serum alt ASTX GSH-PxCorn SOD and MDA contents could be regarded as sensitive indexes to reflect oxidative stress.
