蒙古马高负荷运动训练前后转录组差异表达分析

【摘要】:旨在对蒙古马高负荷运动训练前后转录组差异表达进行分析。本试验对6匹进行了4个月高负荷运动训练的蒙古马进行研究,分别在训练前后两个时期采集肌肉样品,利用二代测序技术对肌肉样品建立转录组文库,对发生显著变化的差异基因进行GO功能富集和KEGG Pathway分析,寻找训练前后发生显著变化的代谢通路及相关候选基因。结果表明,在转录组筛选到的1 102个显著差异表达基因中,有299个在训练后发生下调,803个发生上调。这些差异基因被注释到3 398条GO term上。其中与运动学相关的生物学过程有:肌肉结构发育、心血管系统发育、循环系统发育、肌肉组织发育、肌肉器官发育、肌肉收缩和肌肉细胞分化等。对差异表达基因进行KEGG pathway分析,结果表明,差异基因被富集到如扩张型心肌病、肥厚型心肌病、心肌收缩、钙信号通路、肌动蛋白骨架调节等与运动相关的193条通路中。本研究结果为探究蒙古马强耐力的基本分子机理提供理论基础,同时也为理解蒙古马运动学特性提供新的思路。
[Abstract]:The purpose of this study was to analyze the differential expression of transcriptome in Mongolian horse before and after high-load exercise training. In this experiment, six Mongolian horses trained for 4 months with high-load exercise were studied. Muscle samples were collected before and after training, and the transcriptional library of muscle samples was established by second-generation sequencing technique. Go functional enrichment and KEGG Pathway analysis were performed to identify the metabolic pathways and related candidate genes that changed significantly before and after training. The results showed that 299 of the 1 102 differentially expressed genes were up-regulated after training. These differential genes were annotated to 3 398 go term. The biological processes related to kinematics include muscle structure development, cardiovascular system development, circulatory system development, muscle tissue development, muscle organ development, muscle contraction and muscle cell differentiation. The differentially expressed genes were analyzed by KEGG pathway. The results showed that the differentially expressed genes were enriched into 193 exercise-related pathways such as dilated cardiomyopathy, hypertrophic cardiomyopathy, myocardial contraction, calcium signaling pathway and actin skeleton regulation. The results of this study provide a theoretical basis for exploring the basic molecular mechanism of Mongol horse's strong endurance, and also provide a new idea for understanding the kinematic characteristics of Mongolian horse.
