【摘要】:本试验旨在研究高能低蛋白日粮诱导的蛋鸡脂肪肝脏、肾脏和小肠中自噬、凋亡及相关基因m RNA表达量的变化以及日粮中添加白藜芦醇对其基因表达量的调控作用。144羽海兰褐蛋鸡随机分成正常组(Con)、病理组(HELP)、预防组(HELP+Res)和白藜芦醇组(Con+Res)。试验期为120d,分别于试验第40d、第80d和第120d剖杀12羽蛋鸡,取肝、肾脏及小肠并提取RNA,通过Quantitative Real Time PCR技术检测其ATG5、ATG7、Beclin-1和P62及相关基因Caspase-3、Bax、Bcl-2、HSP60、HSP70、HSP90、apoA-Ⅰ和apoB-100 mRNA表达量。结果如下:1、与Con组比,肝、肾脏及小肠中Beclin-1、ATG5和ATG7 mRNA表达量在HELP组降低,在Con+Res组升高,P62 mRNA则完全相反,均差异极显著(P0.01);与HELP组比,HELP+Res组各组织中Beclin-1、ATG5和ATG7 mRNA表达量升高,P62 m RNA降低,均差异极显著(P0.01)。2、与Con组比,各组织中Caspase-3和Bax mRNA表达量在HELP组升高,在Con+Res组降低,Bcl-2 mRNA则完全相反,均差异极显著(P0.01);与HELP组比,HELP+Res组各组织中Caspase-3和Bax mRNA表达量降低,Bcl-2 mRNA升高,均差异极显著(P0.01)。3、与Con组比,各组织中HSP60、HSP70和HSP90 mRNA表达量在HELP组呈升高,均差异显著(P0.05),在Con+Res组中无明显差异(P0.05);与HELP组,HELP+Res组各组织中HSP60、HSP70和HSP90 mRNA表达量降低,均差异显著(P0.05)。4、与Con组比,各组织apoA-Ⅰ和apoB-100 mRNA表达量在HELP组降低,在Con+Res组升高,均差异极显著(P0.01);与HELP组比,HELP+Res组各组织中apoA-Ⅰ和apoB-100表达量升高,均差异极显著(P0.01)。高能低蛋白诱导的蛋鸡脂肪肝出血综合征会抑制肝、肾脏及小肠自噬、载脂蛋白基因的表达,促进凋亡基和热休克基因的表达,日粮中添加白藜芦醇能对这些基因的表达进行调控。
[Abstract]:The aim of this study was to study the effects of high energy and low protein diets on autophagy in fat liver, kidney and small intestine of laying hens. The changes of mRNA expression of apoptosis and related genes and the regulation of resveratrol on the gene expression in diet. 144 layers were randomly divided into normal group (Con), pathological group (HELP), prevention group (HELP Res) and resveratrol group (Con res), and was divided into three groups: normal group (Con), pathological group (HELP), prevention group (HELP Res) and resveratrol group (Con res).
