日粮对细毛羊瘤胃上皮的SCFA吸收相关基因及氮素转运的调控

【摘要】:本文通过饲养试验、荧光定量和Western blot方法探求日粮不同的NFC/NDF与蛋白水平对内蒙古细毛羊瘤胃上皮VFA吸收相关基因表达量及氮素转运的调控。本论文的研究内容和结果具体如下:试验1日粮不同NFC/NDF对细毛羊消化代谢、瘤胃内环境及SCFA吸收的影响选用9只体况良好,体重为43.8±4.75Kg有永久性瘤胃瘘管的内蒙古鄂尔多斯细毛羊半同胞羯羊,按体重分为3组:NFC/NDF分别为:0.35、0.66和1.14,蛋白水平相同,为13.5%。饲养试验表明日粮NFC/NDF对细毛羊氮代谢无显著影响。粪中的NDF随日粮NFC/NDF的增加而减少。瘤胃总VFA随日粮NFC/NDF的增加而增加,其中乙酸的浓度随NFC/NDF增加而降低,丙酸和丁酸的浓度增加。分子实验结果表明:各组间MCT1、PAT1、 vH+ATPase等基因的表达量没有显著影响(P0.05)。NHE3和DRA的表达量随日粮NFC的增加而增加。MCT4的表达量反而随日粮NFC的增加而降低。NHE1和Na+、K+ATPase的表达量在NCF/NDF为0.66时最高,而NFC/NDF过低或过高都会降低其表达量。试验2日粮不同NFC/NDF和RDP对对细毛羊消化代谢、瘤胃内环境及SCFA吸收和氮素转运的影响选用12只体况良好,体重为44.2±4.35Kg有永久性瘤胃瘘管的内蒙古鄂尔多斯细毛羊半同胞羯羊,按体重分为4组:1组,NFC/NDF:1.14, CP:15.5:2组,NFC/NDF:1.14,CP:10.5;3组NFC/NDF:0.35,CP:15.5:4组,NFC/NDF:0.35,CP:10.5。饲养管理同试验l。结果表明,细毛羊氮代谢受日粮RDF的影响,随日粮RDP的增加而增加。而各组间粪氮和粪NDF含量无显著差异。瘤胃PH随日粮NFC/NDF的增加而降低。瘤胃NH3-N的浓度随日粮RDP的增加而增加,并且日粮NFC/NDF低时NH3-N浓度明显增加。日粮NFC/NDF和RDP对NHE1、Na+、K+ATPase和H+ATPase的表达量无显著影响(P0.05)。NFC/NDF和RPD都相对较高时,MCTl和MCT4的表达量最高。NHE3的表达量随NFC/NDF的增加而增加,受日粮RDP的影响。日粮NFC/NDF相同时,低N日粮可增加UT-B的表达量。日粮蛋白水平相同时,低NFC/NDF组的UT-B表达量增加。日粮NFC/NDF为较高(1.14)时,水通道蛋白的表达量随日粮蛋白的增加而降低,而日粮NFC/NDF为较低(0.35)时,水通道蛋白的表达量随日粮蛋白的增加而增加。日粮NFC/NDF为较高(1.14)时,氨转运蛋白的表达随日粮蛋白的增加而降低,日粮NFC/NDF为较低(0.35)时,氨转运蛋白表达量随日粮蛋白的增加而增加。试验3限饲及之后的恢复对瘤胃内环境和血液生化指标的影响试验动物、分组和饲养管理同试验1。结果表明,总VFA的浓度在63.68-89.49mmol/L之间,随日粮NFC/NDF的增加而增加。乙酸的浓度在48.35-65.75之间mmol/L,随日粮NFC/NDF的增加而减少。除11:00以外,乙酸和丙酸浓度各组之间无显著差异。各组间NH3-N的浓度无显著差异(P0.05)。而pH只随日粮NFC/NDF的增加而显著降低。限饲之后突然的恢复日粮将破坏瘤胃内环境,使瘤胃pH降低。
[Abstract]:The regulation of VFA absorption related gene expression and nitrogen transport in ruminal epithelium of Inner Mongolia fine wool sheep was investigated by feeding experiments, fluorescence quantitative and Western blot methods. The contents and results of this paper were as follows: the digestion and metabolism of different NFC/NDF to fine wool sheep in the 1 day diet and the rumen in the rumen were tested in this paper. The effects of environment and SCFA absorption were divided into 9 half sib sheep of Inner Mongolia Ordos fine wool sheep with a good body weight of 43.8 + 4.75Kg and a permanent rumen fistula. The expression of K+ATPase was the highest when NCF/NDF was 0.66, while NFC/NDF was too low or too high to reduce its expression. When dietary NFC/NDF was higher (1.14), the expression of water channel protein decreased with the increase of dietary protein, while the expression of water channel protein increased with the increase of dietary protein when NFC/NDF was lower (0.35). When dietary NFC/NDF was higher (1.14), the expression of ammonia transporter decreased with the increase of dietary protein, and NFC/NDF in diet. At lower (0.35), the expression of ammonia transporter increased with the increase of dietary protein. Experimental animals, group and feeding management and test 1. showed that the effects of 3 limited feeding and recovery on the rumen environment and blood biochemical indexes showed that the concentration of total VFA was between 63.68-89.49mmol/ L and increased with the increase of dietary NFC/NDF. The concentration of mmol/L decreased with the increase of NFC/NDF in the diet. There was no significant difference between the concentrations of acetic acid and propionic acid except at 11:00. There was no significant difference between the concentrations of acetic acid and propionic acid. The concentration of NH3-N in each group was not significantly different (P0.05). But the pH only decreased with the increase of NFC/NDF in the diet. The ruminal pH was destroyed by the sudden recovery of the rumen environment after the limited feeding. Lower.
