奶牛酮病检测方法的比较和牛乳、血清中β-羟丁酸相关性研究
[Abstract]:Ketosis is a common metabolic disorder caused by carbohydrate and fat metabolism in dairy cows. It is characterized by ketonemia, ketouria and ketone lactation. It is characterized by hypoglycemia, increased free fatty acids in blood and fatty liver. Glycogen level decreased, mainly in high-yielding cows. Although ketosis can be cured and rarely causes cows to die, ketosis can lead to a decrease in milk production, a decrease in milk quality and reproduction, secondary diseases such as reproductive system diseases and endocrine disorders, which increase the cost of treatment. To the dairy cattle industry caused a large loss. Timely and effective prevention and treatment of ketosis in dairy cattle is of great significance to improve the development level of dairy cattle breeding industry. Objective: to determine the concentration of ketone and 尾 -hydroxybutyric acid in milk and serum of 96 dairy cows in Shihezi area. The aim of this study was to study the relationship between ketosis monitoring methods and 尾 -hydroxybutyric acid (BHBA) concentrations in milk and serum of dairy cows. Methods the milk was detected qualitatively by the methods of 1: 1, ketone powder, ross and Rothera, and the serum of cows was detected by modified salicylaldehyde method. The ketone body content in serum was defined as ketosis cow. The 尾 -hydroxybutyric acid concentration in milk was determined by enzyme fluorescence method after deproteinization of ketosis cow milk by phosphotungstic acid precipitation method. Serum 尾 -hydroxybutyric acid (尾 -hydroxybutyric acid) concentration in ketosis cows was determined by enzyme-linked immunosorbent assay (Elisa) and analyzed by SPSS 10.0. The correlation coefficient was Pearson. Results by using the modified salicylaldehyde method, 40 cows were found to suffer from ketosis, and the accuracy of ketone powder method was the highest among the four qualitative methods. The coincidence rate was 85.42 路2.The average concentration of BHBA in milk of ketosis cow was 1.21 卤0.25 mmol / L, and the average concentration of BHBA in serum was 2.19 卤0.35 mmol / L. The concentration of BHBA in milk gradually increased with the increase of birth number, but the concentration of BHBA in serum did not change significantly, and the correlation coefficient between BHBA and BHBA was higher. The higher the milk yield, the higher the BHBA content in milk and serum, and the correlation coefficient was higher. Conclusion the detection efficiency and false positive rate of ketone powder method are high and false positive rate is low. The correlation between milk and serum 尾 -hydroxybutyric acid is 0.8102-0.9105, so it can be determined whether it is ketosis cow or not by detecting 尾 -hydroxybutyric acid in milk, which can be used as the basis for early diagnosis of ketosis in dairy cows.
