1例鸡心包积水-肝炎综合征的实验室诊断

【摘要】:2015年9月份,江苏省某养殖场饲养的肉鸡发生了一种传染病,发病鸡以为精神沉郁、潜伏期短、病死率高为特征;剖检可见心包积液和肝脏呈土黄色并分布有出血点。无菌采集病鸡的肝脏和心包积液,分别用麦康凯琼脂平板和血平板进行细菌分离,结果均为阴性。根据包涵体肝炎(IBH)的Hexon基因设计引物用于该病原体的检测,结果显示,以病鸡的肝脏和心包积液提取的DNA为模板,通过PCR可以扩增出约500 bp大小的IBH特异性条带,经序列测定及分析,判定为FAd V-4型腺病毒。通过实验室诊断技术结合临床症状和流行病学等进行综合分析,将此次疫病确诊为FAd V-4型腺病毒感染所致。
[Abstract]:In September 2015, an infectious disease occurred in broilers raised at a farm in Jiangsu Province. The diseased chickens were characterized by depression, short incubation period and high mortality, and the pericardial effusion and liver were found to be yellowish and haemorrhage. The liver and pericardial effusion of diseased chickens were collected and isolated by wheat Kang Kai Agar plate and blood plate respectively. The results were negative. According to the Hexon gene of inclusion body hepatitis (IBH), primers were designed to detect the pathogen. The results showed that using DNA extracted from liver and pericardial effusion of diseased chicken as template, about 500bp IBH specific bands could be amplified by PCR. FAd V-4 adenovirus was identified by sequencing and analysis. The disease was diagnosed as a result of FAd V-4 adenovirus infection through laboratory diagnosis combined with clinical symptoms and epidemiology.
