贵州地方山羊ERα多态性及组织差异表达

【摘要】:雌激素受体α(estrogen receptor,ERα)基因在动物繁殖过程中起着重要作用,已经被认为是影响猪(Sus scrofa)产仔数的主效基因,但在山羊(Capra hicus)方面研究较少。为了揭示雌激素受体基因对贵州本地山羊繁殖性状的影响,本研究采用直接测序技术检测了该基因8个外显子的SNP,并使用qRT-PCR检测ERα基因在黔北麻羊的下丘脑,垂体,性腺轴中不同表达量。结果表明,ERα基因在黔北麻羊和贵州黑山羊第8外显子中均存在C127T突变,且属于错义突变,致使该基因mRNA二级结构以及编码蛋白结构的改变。在第8外显子的C127T位点发现CC、CT和TT 3种基因型,其中,在黔北麻羊群体中CC基因型个体的产羔数与TT和CT基因型间达差异显著水平(P0.05),在贵州黑山羊群体中未达到显著差异。qRT-PCR检测结果显示,单羔组和多羔组黔北麻羊的5个组织中ERα基因均有表达,且卵巢均最高。进一步研究发现,单羔组和多羔组黔北麻羊卵巢组织中的ERαmRNA表达量差异显著(P0.05),而在其他组织间均未达到差异显著(P0.05)。结果显示,ERα中的C127T突变可能影响贵州地方山羊的产羔数,为培育贵州地方山羊高繁殖力品系提供了理论依据。
[Abstract]:Estrogen receptor ER 伪 (ER 伪) gene plays an important role in animal reproduction. It has been considered as a major gene affecting piglet (Sus scrofa) litter size, but little research has been done on goat (Capra hicus). In order to reveal the effect of estrogen receptor gene on reproductive traits of Guizhou native goats, the SNPs of 8 exons of the gene were detected by direct sequencing, and the ER 伪 gene was detected by qRT-PCR in the hypothalamus and pituitary of Qianbei Ma sheep. Different levels of expression were found in the gonadal axis. The results showed that there was C127T mutation and missense mutation in exon 8 of Qianbei Ma sheep and Guizhou black goat, which resulted in the change of mRNA secondary structure and coding protein structure. Three genotypes of CCT CT and TT were found at the C127T locus of exon 8, in which, There was significant difference in lambs size among CC genotype individuals and TT and CT genotypes (P0.05), but there was no significant difference in Guizhou black goat population by qRT-PCR. The ER 伪 gene was expressed in 5 tissues of Qianbei Ma sheep in single lambs and many lambs, and the ovary was the highest. Further study showed that the expression of ER 伪 mRNA in ovarian tissues of single lambs group and multiple lambs group was significantly different (P0.05), but there was no significant difference among other tissues (P0.05). The results showed that the C127T mutation in ER 伪 might affect the lambing size of Guizhou local goats, which provided a theoretical basis for breeding Guizhou local goat lines with high fecundity.
