精子不同处理方式对绵羊胞质内单精子注射效率的影响

【摘要】:胞质内单精子注射前对精子进行预处理是模拟精子受精过程的变化增强精子解聚能力。常用的精子处理方法有聚乙烯吡咯烷酮、二硫苏糖醇、冻融处理等。研究表明ICSI卵的卵裂率低的主要原因是精子解聚能力差。因此研究精子的处理方法提高ICSI的注射效率是十分必要的。该实验探讨了精子不同预处理方法对ICSI胚胎发育的影响。利用超声波细胞粉碎仪对精子做不同时间(3 s、6 s)的处理,观察ICSI胚胎发育情况,选择最佳的处理时间,然后利用超声波分别处理鲜精和冻精,比较不同精液对ICSI胚胎发育的影响。将精子分别利用超声波和碱处理,对比两种不同预处理方法对ICSI卵卵裂率和桑囊胚率的影响,结果表明:(1)超声波处理绵羊精子最佳的处理时间是6 s;(2)鲜精和冻精经超声波处理后对ICSI卵的卵裂率(39.1%vs 37.4%,P0.05)和桑囊率(12.3%vs 13.2%,P0.05)影响不显著;(3)冻精经碱处理后其ICSI卵的卵裂率(46.6%vs37.4%,P0.05)和桑囊胚率(20.4%vs 13.2%,P0.05)均显著高于超声波处理冻精。
[Abstract]:Pretreatment of spermatozoa before intracytoplasmic sperm injection is to simulate the change of sperm fertilization process to enhance the ability of sperm depolymerization. Common sperm treatment methods are polyvinylpyrrolidone, dithiothrelitol, freeze-thaw treatment and so on. The main reason for the low cleavage rate of ICSI eggs is the poor ability of sperm depolymerization. Therefore, it is necessary to study the sperm processing method to improve the injection efficiency of ICSI. The effects of different sperm pretreatment methods on the development of ICSI embryos were studied. Spermatozoa were treated with ultrasonic cell pulverizer for different time (3 s / 6 s). The development of ICSI embryos was observed and the best treatment time was selected. Then the fresh and frozen spermatozoa were treated with ultrasonic wave, respectively. The effects of different semen on ICSI embryo development were compared. The sperm were treated with ultrasound and alkali respectively. The effects of two different pretreatment methods on the cleavage rate and blastocyst rate of ICSI were compared. The results showed that: (1) the best treatment time of ultrasonic treatment was 6 s; (2) the cleavage rate of ICSI eggs (39.1%vs 37.4%) and mulberry sac rate (12.3%vs 13.2P 0.05) were not significantly affected by ultrasonic treatment of fresh and frozen spermatozoa; (3) the cleavage of ICSI eggs of frozen spermatozoa treated with alkali was not significant. The rates (46.6% vs 37.4%) and mulberry blastocyst rate (20.4%vs 13.2%) were significantly higher than that of ultrasonic treatment.
