奶牛妊娠黄体蛋白组学研究

【摘要】：为了研究奶牛黄体的形成及退化过程的相关机理,并进一步探究更加准确的妊娠及相关生理过程的蛋白标记物,试验采用蛋白质组学经典的二维凝胶电泳(2-DE)技术检测和分析妊娠前后黄体组织间的差异蛋白,使用PDQuest8.0软件检测,取差异蛋白点进行分析,并经MALDI TOF/TOFMS/MS检测。结果表明:6种差异蛋白分别为26s蛋白酶体蛋白、纤维蛋白、DNA拓扑异构酶蛋白、核纤层蛋白、硫氧还原蛋白、微管蛋白。对这6种蛋白在组织间的表达差异进行分析,发现这些蛋白可能与黄体形成和黄体退化及黄体激素分泌有关。
[Abstract]:In order to study the mechanism of luteal formation and degeneration in dairy cows, and to explore more accurate protein markers of pregnancy and related physiological processes, Two dimensional gel electrophoresis (2-DE), a classical proteomics technique, was used to detect and analyze differentially expressed proteins in corpus luteum before and after pregnancy. PDQuest8.0 software was used to detect the differential proteins and MALDI TOF/TOFMS/MS was used to detect them. The results showed that the six differential proteins were 26s proteasome protein, fibrin DNA topoisomerase protein, nuclear laminar protein, thioxy-reducing protein and tubulin. It was found that these proteins might be related to luteal formation, luteal degeneration and luteal hormone secretion.
