蛋鸭极低密度脂蛋白受体基因多态性与蛋品质相关性分析

【摘要】:极低密度脂蛋白受体基因(VLDLR)在卵生动物蛋黄物质形成过程起关键作用。研究以国绍Ⅰ号母鸭为研究对象,采用一代测序法对VLDLR基因外显子12至外显子13区域进行SNPs筛选,并分析各多态位点间的连锁不平衡状态及不同基因型与蛋品质的相关性。结果显示:共发现4个多态位点,其中S1(A7120G)位于12号外显子,未引起氨基酸序列改变;S2(A7261G)位于12号内含子;S3(C7325T)和S4(A7337G)位于13号外显子中,分别导致Thr突变为Met,Asn突变为Asp。位点S1、S2和S4为强连锁不平衡位点,存在3种单倍型和3种双倍型,单一位点基因型和双倍型与蛋黄比例、蛋重、蛋黄重、蛋壳重和蛋黄颜色的相关性分析结果均为不显著相关,在国绍Ⅰ号VLDLR基因外显子12-外显子13区域内未发现与蛋黄比例等蛋品质相关的分子标记SNPs位点。
[Abstract]:Very low density lipoprotein receptor (VLDLR) gene plays a key role in egg yolk formation. In this study, the first generation sequencing method was used to screen the region of exon 12 to exon 13 of VLDLR gene, and the linkage disequilibrium state among the polymorphic loci and the correlation between different genotypes and egg quality were analyzed. The results showed that four polymorphic loci were found, of which S1 (A7120G) was located in exon 12. No amino acid sequence change was observed. S2 (A7261G) was located in exon 13 of intron 12, and S4 (A7337G) was located in exon 13, respectively, resulting in the mutation of Thr into Asp.. The results showed that there were three haplotypes and three diplotypes. The correlation analysis of single locus genotype and diplotype with yolk ratio, egg weight, egg yolk weight, egg shell weight and yolk color was not significant. No SNPs locus related to egg quality such as yolk ratio was found in exon 12- exon 13 of VLDLR gene of GuoShao 鈪,
