替代饲用抗生素的复方中草药提取物抗氧化功能研究

【摘要】:本文以课题组前期研制的替代饲用抗生素的复方中草药提取物为原料,研究该提取物及其组成的各单味药的体外清除自由基功能、体内抗氧化功能,并对其抗氧化成分进行初步测定分析,探索其抗氧化机制,为进一步开发利用饲用抗生素的替代物提供理论依据。1复方中草药提取物清除自由基活性研究。通过体外抗氧化试验研究复方中草药提取物及其组成的各单味药对DPPH·自由基、羟自由基的清除作用,并测定了其还原能力。结果显示:复方中草药提取物及其各单味药对DPPH·自由基、羟自由基均具有较强的清除能力,但同一种药对不同自由基的清除效果不同。其中,对DPPH·自由基的清除作用强弱顺序为:五倍子复方1香薷野菊花乌梅大青叶复方2柴胡;对羟自由基的清除作用强弱顺序为:五倍子乌梅复方1复方2大青叶野菊花香薷柴胡;在所测定浓度范围内,复方中草药提取物及其各单味药还原能力随浓度的增加而增大。在相同浓度下,中草药提取物的还原能力大小顺序为:五倍子复方1野菊花香薷乌梅复方2大青叶柴胡。2复方中草药提取物体内抗氧化活性研究。通过动物试验研究以乌梅为主要成分的复方中草药提取物对小鼠外周血、心脏、肝脏、脾脏、肾脏中抗氧化酶SOD、GSH-PX活力及脂质过氧化物MDA含量的影响。结果显示:复方2对小鼠机体具有较强的抗氧化能力。该提取物在100、200、400mg/kg·bw三个剂量水平上均可导致小鼠外周血、心、肝、脾、肾组织中SOD和GSH-Px活性显著增强,降低小鼠上述组织中MDA含量,并呈现剂量反应关系。其中,在200mg/kg·bw剂量下,上述组织SOD活性依次提高了46.44%、26.55%、55.15%、34.71%、83.74%,GSH-PX活性依次提高了17.23%、0.58%、44.57%、49.54%、22.49%,MDA含量依次下降了13.65%、34.42%、11.60%、16.63%、28.73%。3复方中草药提取物抗氧化活性成分分析。通过紫外显色反应测定各复方中草药提取物及其各单味药中多酚及黄酮的含量。结果显示:复方中草药提取物及其各单味药中多酚含量从高到低依次为:五倍子复方1香薷复方2野菊花乌梅大青叶柴胡,黄酮含量从高到低依次为:为五倍子香薷复方1复方2野菊花乌梅大青叶柴胡。其中五倍子总酚和黄酮含量最高,分别为30.59ug/mg、58.47ug/mg。复方中草药提取物及其各单味药对DPPH·自由基IC50值与总酚含量的相关系数r=-0.422;与黄酮含量的相关系数r=-0.609。复方中草药提取物及其各单味药对·OH自由基IC50值与总酚含量的相关系数r=-0.443;与黄酮含量的相关系数r=-0.506。由复方中草药提取物及其各单味药对DPPH·自由基、·OH自由基的IC50值与总酚或黄酮含量的相关系数可知,其对DPPH·和·OH自由基呈中度相关,说明除了多酚、黄酮外,中草药对自由基清除作用还与其所含的其他成分有关。结论:复方中草药提取物及其各单味药对DPPH·自由基、羟自由基均具有较强的清除能力,在所测定浓度范围内,复方中草药提取物及其各单味药还原能力随浓度的增加而增大;复方2对小鼠机体具有较强的抗氧化能力;复方中草药提取物及其各单味药具有清除自由基的作用与其多酚、黄酮的含量呈中度相关。
[Abstract]:In this paper, the compound Chinese herbal medicine extract, which was developed by our research group as a substitute for feeding antibiotics, was used as raw material to study the free radical scavenging function in vitro and antioxidant function in vivo of the extract and its constituents. The antioxidant components were determined and analyzed preliminarily, and the antioxidant mechanism was explored to further develop and utilize feeding antibiotics. The scavenging effect of compound Chinese herbal extracts and their constituents on DPPH free radicals and hydroxyl free radicals was studied by antioxidant test in vitro, and their reducing ability was determined. The scavenging effect of the same drug on DPPH free radicals was different. The order of scavenging effect on DPPH free radicals was as follows: Galla Chinensis Compound 1 Fragrant Elsholtzia Wild Chrysanthemum Flower, Mume and Daqing Leaf Compound 2 Bupleurum; the order of scavenging effect on hydroxyl radicals was as follows: Galla Chinensis Blume. Compound 1, Compound 1, Compound 2, Compound 1, Compound 1, Compound 2, Compound 2, Compound 2, Compound 2, Compound 2, Compound 2, Compound 2, Compound 2, Compound 2, Compound 2, Compound Chinese Herbal Medicine, Compound 1, Compound 1, Compound 1, Compound 1, Compound 2, Compound 2, Compound 2, Compound 2, Compound 2, Compound 2, Compound Chinese Herbal Medicine, Compound 1, Compo The antioxidant activity of the extract in vivo was studied.The antioxidant enzyme activities of SOD,GSH-PX and the content of MDA in peripheral blood,heart,liver,spleen and kidney of mice were studied by animal experiment.The results showed that compound 2 had strong antioxidant activity in mice. The activity of SOD and GSH-Px in peripheral blood, heart, liver, spleen and kidney tissues of mice were significantly increased at 100,200,400 mg/kg/bw levels, and the MDA content in the above tissues of mice was decreased in dose-response relationship. The SOD activity in the above tissues was increased by 46.44%, 26.55%, 55.15%, 34.71%, 83.71% at 200 mg/kg/bw dosage. The activity of GSH-PX increased by 17.23%, 0.58%, 44.57%, 49.54%, 22.49%, and the content of MDA decreased by 13.65%, 34.42%, 11.60%, 16.63% and 28.73% respectively. 3 Compound Chinese herbal extracts were analyzed for their antioxidant activity components. The content of polyphenols and flavonoids in each compound Chinese herbal extract and its single herbal medicine was determined by UV reaction. The contents of polyphenols in the extracts of Chinese herbal medicines and their single herbs were as follows: Galla Chinensis Compound Prescription 1 Xiangqiao Compound Prescription 2 Wild Chrysanthemum Black Plum Daqing Leaf Bupleurum. The contents of flavonoids were as follows: Galla Chinensis Compound Prescription 1 Wild Chrysanthemum Compound Prescription 2 Wild Chrysanthemum Black Plum Blue Leaf Bupleurum. 47ug/mg. The correlation coefficient between the IC50 value of DPPH radical and the content of total phenols of the extracts of Chinese herbal medicines and their single herbs was r = - 0.422; the correlation coefficient with the content of flavonoids was r = - 0.609. The correlation coefficient between the IC50 value of OH radical and the content of total phenols of the extracts of Chinese herbal medicines and their single herbs was r = - 0.443; the correlation coefficient with the content of flavonoids was r = - 0.506. The correlation coefficients between the IC50 values of DPPH free radicals and OH free radicals and the contents of total phenols or flavonoids showed that the IC50 values were moderately correlated with DPPH and OH free radicals, indicating that the scavenging effect of Chinese herbal medicine on free radicals was related to other components besides polyphenols and flavonoids. The extracts and their individual medicines have strong scavenging capacity on DPPH free radicals and hydroxyl free radicals. Within the range of concentration determined, the reducing capacity of compound Chinese herbal extracts and their individual medicines increases with the increase of concentration; compound 2 has strong antioxidant capacity in mice; compound Chinese herbal extracts and their individual medicines have strong antioxidant capacity. The scavenging effect of free radicals was moderately related to polyphenols and flavonoids.
