多型口蹄疫疫苗灭活时间和纯化的比较研究

【摘要】:【目的】为了探讨不同灭活时间及纯化对多型口蹄疫疫苗146 S、总蛋白量、抗原收获量及疫苗效力的影响。【方法】选取OM/Re-A/AF/Asia1 4种不同型的口蹄疫疫苗为研究对象,设定不同灭活时间(8、10、12、14和16 h),对每个时间点疫苗母液中146 S、总蛋白和抗原收获量进行检测和差异性分析;测量纯化前后146 S和总蛋白的变化趋势;测定不同灭活时间及纯化前后疫苗PD50数值进行判定效力。【结果】灭活不同时间点,146 S含量和抗原收获量随着时间先增高后平缓的趋势,146 S在14 h出现平缓,而抗原收获量在10 h时开始平缓;146 S含量OM和AF型明显高于Re-A和Asia1型,抗原收获量Asia1型最高;总蛋白含量随着时间呈现先降低后平缓的趋势,AF型口蹄疫疫苗降低最明显;疫苗纯化前后相比分析发现,146 S含量明显升高,总蛋白含量明显降低(P0.01)。4种疫苗PD50值随着纯化时间先增大后平稳,纯化后PD50显著提高(P0.01),免疫效力增强。【结论】OM/Re-A/AF/Asia1 4种不同型的口蹄疫疫苗在纯化不同时间点,疫苗中146 S、抗原收获量和总蛋白含量呈规律性变化,且纯化前后146 S、总蛋白含量及PD50值差异性极显著,表明口蹄疫疫苗的灭活时间和纯化是影响疫苗效价的重要因素。
[Abstract]:[objective] to investigate the effects of different inactivation time and purification on the amount of protein, total protein, antigen harvest and vaccine effectiveness of multiform foot-and-mouth disease vaccine 146S. [methods] four different types of foot-and-mouth disease vaccine (OM/Re-A/AF/Asia1) were selected as the research object. Different inactivation time (810) 12C 14 and 16 h), were set to detect and analyze the yield of 146s, total protein and antigen in the maternal liquor of vaccine at each time point, and the change trend of 146S and total protein before and after purification were measured. [results] the trend of inactivation of 146S and antigen harvest at different time points was that the level of 146S and then the yield of antigen increased first and then slowly at 14 h after inactivation, the efficacy was determined by measuring the different inactivation time and the PD50 value of the vaccine before and after purification. However, the OM and AF types were significantly higher than those of Re-A and Asia1 at 10 h, and the total protein content was the highest in Asia1 type, and the total protein content decreased first and then slowly. The decrease of AF type foot-and-mouth disease vaccine was the most obvious. Compared with those before and after purification, the results showed that the content of total protein was significantly decreased (P0.01), and the PD50 value of the vaccine increased first and then stabilized with the purification time. After purification, PD50 was significantly increased (P0.01), and the immune potency was enhanced. [conclusion] at different time points of purification, 146s, antigen harvest and total protein content of OM/Re-A/AF/Asia1 four different types of FMD vaccine showed regular changes. There were significant differences in total protein content and PD50 before and after purification, indicating that the inactivation time and purification of foot-and-mouth disease vaccine were important factors affecting the titer of the vaccine.
