银杏外种皮多糖的制备及其抗新城疫病毒作用的初步研究

【摘要】:为了探索银杏外种皮多糖(GBEP)在鸡胚中对新城疫病毒(NDV)的作用,试验采用水提醇沉法获得多糖(含量为51.9%),然后用不同浓度多糖接种9~11日龄鸡胚测定多糖的安全浓度,在此基础上比较不同给药方式(先给药再攻毒、先攻毒再给药及药物和病毒混合孵育后再攻毒)对NDV的作用。结果表明:GBEP在不高于4 mg/枚鸡胚时不影响鸡胚的正常发育。0.8 mg/枚GBEP处理组的胚体发育、HA血凝价均与利巴韦林组相近,而攻毒对照组鸡胚在48小时时全部死亡。先攻毒再给药组鸡胚在48小时时出现5枚死亡,60小时时全部死亡;先给药再攻毒组鸡胚在同时期出现1枚死亡,而药物与病毒混合孵育后再攻毒组未出现死亡,且72小时时胚体长度明显长于攻毒对照组(P0.05)。说明GBEP具有明显的预防和直接抗NDV感染鸡胚的作用。
[Abstract]:In order to explore the effect of Ginkgo biloba outer coat polysaccharide (GBEP) on Newcastle disease virus (NDV) (NDV) in chicken embryo, the polysaccharide was obtained by water extraction and alcohol precipitation (51.9%), and the safe concentration of polysaccharides was determined by inoculating 9 ~ 11 day-old chicken embryos with different concentrations of polysaccharides. On this basis, the effects of different drug administration methods (first drug administration and then drug attack, first attack on poison and then drug and virus mixed incubation and then attack on virus) on NDV were compared. The results showed that the normal development of chicken embryos was not affected by GBEP when the number of chicken embryos was not more than 4 mg/. The embryo body development of 0.8 mg/ GBEP treatment group was similar to that of ribavirin group, while that of control group died at 48 hours. Five chicken embryos died at 48 hours and all died at 60 hours in the first attack group, and one embryo died in the same period in the first administration group, but there was no death in the second attack group after the drug and virus were incubated. The length of embryo body at 72 hours was significantly longer than that of control group (P0.05). The results showed that GBEP could prevent and directly resist NDV infection in chicken embryos.
