猪流行性腹泻病毒河南株2014年~2015年分子特征和遗传进化分析

【摘要】:为分析河南省猪流行性腹泻病毒(PEDV)分子和遗传进化特征,本研究对2014年1月~2015年12月收集自河南省不同地区的200份PEDV疑似病料进行RT-PCR检测,并对15份阳性样品进行S和ORF3基因的克隆及序列分析,结果显示15株河南流行株PEDV的S基因和ORF3基因的部分核苷酸和氨基酸序列与参考株相比均发生了变异。S和ORF3基因遗传进化树显示,PEDV均可分为两个群,基于S基因的进化树显示15株河南株独立成群,与其它参考株亲缘关系较远;基于ORF3基因的进化树显示,15株河南株与国内分离株、美国株及韩国株均有较近的亲缘关系。从S和ORF3基因的进化关系中还显示,15株河南株与经典株CV777及国内所使用的疫苗株,亲缘关系较远。表明PEDV河南株有变异和进化的趋势。本研究为更好地控制本病在河南地区的发生奠定基础。
[Abstract]:In order to analyze the molecular and genetic characteristics of porcine epidemic diarrhea virus (PEDV) in Henan Province, 200 suspected PEDV samples collected from different areas of Henan Province from January 2014 to December 2015 were detected by RT-PCR. The S and ORF3 genes were cloned and sequenced from 15 positive samples. The results showed that the nucleotide and amino acid sequences of the S gene and the ORF3 gene of 15 Henan epidemic strains were mutated compared with the reference strains. The S and ORF3 gene genetic evolution tree showed that PEDV could be divided into two groups. The phylogenetic tree based on S gene showed that 15 Henan strains were independent in groups and had a close relationship with other reference strains, while the evolutionary tree based on ORF3 gene showed that 15 Henan strains were closely related to domestic isolates, American strains and Korean strains. From the evolutionary relationship of S and ORF3 genes, 15 Henan strains were closely related to the classical strain CV777 and the vaccine strains used in China. The results indicated that PEDV Henan strain had the tendency of variation and evolution. This study lays a foundation for better control of the disease in Henan province.
