金丝桃苷对小鼠T淋巴细胞亚群及血清细胞因子的影响

【摘要】:观察金丝桃苷对小鼠免疫系统的作用,将Balb/c小鼠随机分成5组,空白对照组(在第1、3、5天腹腔注射生理盐水)、免疫抑制组(在第1、3、5天腹腔注射环磷酰胺)、给药组(在第1、3、5天腹腔注射环磷酰胺,第1~7天每天以150/300mg/kg金丝桃苷水溶液灌胃)和阳性对照组(在第1、3、5天腹腔注射环磷酰胺,第1~7天每天以0.2mg/kg黄芪多糖水溶液灌胃)。用ELISA法测定小鼠T淋巴细胞亚群CD_3、CD_4、CD_8以及细胞因子IL-4、IL-6、IL-10、IFN-γ、TNF-α的水平。结果表明,与空白对照组相比,免疫抑制组的CD_3、CD_4、CD_8水平以及血清细胞因子IFN-γ、TNF-α水平显著降低(P0.05),IL-4、IL-6及IL-10水平显著升高(P0.05)。给予金丝桃苷后第8天,与免疫抑制组相比,CD_3、CD_4、CD_8水平以及血清细胞因子IFN-γ、TNF-α水平极显著升高(P0.01),IL-4、IL-6、IL-10水平极显著降低(P0.01)。金丝桃苷能通过提高免疫抑制小鼠T淋巴细胞亚群的CD_3、CD_4、CD_8水平以及细胞因子IFN-γ及TNF-α水平,降低IL-4、IL-6、IL-10水平,来拮抗环磷酰胺所致的免疫机能抑制,提高机体免疫功能。
[Abstract]:To observe the effect of hyperin on the immune system of mice, Balb/c mice were randomly divided into 5 groups. The blank control group (intraperitoneal injection of normal saline on the 1st day 3 days), the immunosuppressive group (intraperitoneal injection of cyclophosphamide on the 1st day 3 days), the administration group (intraperitoneal injection of cyclophosphamide on the 1st day 3 days), the immunosuppressive group (intraperitoneal injection of cyclophosphamide on the 1st day 3 days), 150/300mg/kg hyperoside solution was administered daily on day 1 to day 7, and the positive control group was given cyclophosphamide intraperitoneally on day 1 and day 7 respectively. 0.2mg/kg Astragalus membranaceus polysaccharide solution was administered intragastrically on day 1 and day 7 respectively. The levels of CD_3,CD_4,CD_8 and cytokines IL-4,IL-6,IL-10,IFN- 纬 and TNF- 伪 in T lymphocyte subsets of mice were determined by ELISA method. The results showed that compared with the control group, the levels of CD_3,CD_4,CD_8 and serum cytokines IFN- 纬 and TNF- 伪 in immunosuppressive group were significantly decreased (P0.05), and the levels of IL-4,IL-6 and IL-10 were significantly increased (P0.05). On the 8th day after hypericin administration, the levels of CD_3,CD_4,CD_8, serum cytokines IFN- 纬 and TNF- 伪 were significantly increased (P0.01) and IL-4,IL-6,IL-10 levels were significantly decreased (P0.01) compared with the immunosuppressive group. Hypericin can antagonize the immunosuppression of cyclophosphamide by increasing the level of CD_3,CD_4,CD_8 of T lymphocyte subsets, the levels of cytokines IFN- 纬 and TNF- 伪, and decreasing the level of IL-4,IL-6,IL-10, which can antagonize the inhibition of immune function induced by cyclophosphamide and improve the immune function of the body.
