2014年甘肃省三个地区绵羊布氏杆菌病的流行病学调查

【摘要】:布氏杆菌病(Brucellosis),简称布病,是一种由布氏杆菌(Brucella)引起的人兽共患传染病。近年来,甘肃省的绵羊布病疫情呈现上升趋势,不仅制约绵羊养殖业的持续发展,而且影响人民群众的身体健康。本论文对2014年甘肃省三个地区绵羊布病疫情进行调查。1.在无“金标准”情况下,为评价RBT和I-ELISA对绵羊布病的诊断能力,本研究用RBT和I-ELISA对采自某地的176份未免疫绵羊血清同时进行检测,通过建立贝叶斯模型,计算两种方法的灵敏度和特异度。结果显示,RBT和I-ELISA的灵敏度分别为76.5%、89.6%,特异度分别为95.1%、93.4%,I-ELISA的诊断能力更好。2.在靖远县、崆峒区和永昌县的54个养殖场采集血清1536份,经I-ELISA检测,个体总阳性率为3.58%(55/1536),群体总阳性率为25.93%(14/54)。靖远县、崆峒区和永昌县的个体阳性率分别为3.35%(18/538)、6.02%(30/498)和1.40%(7/500);群体阳性率分别为22.22%(2/9)、33.33%(11/33)和8.30%(1/12)。3.采集流产胎儿病料,用布氏杆菌培养基分别在靖远县、崆峒区和永昌县分离得到1株布氏杆菌,依次命名为GS0250、GS2620和GS2638。用传统方法、AMOS-PCR和MLVA-16对3株布氏杆菌进行种型鉴定。传统方法将3株菌均鉴定为羊种3型布氏杆菌、AMOS-PCR扩增均得到731bp的特异性片段,将其鉴定为羊种布氏杆菌。3株菌的MLVA型为1-5-3-13-2-2-3-2-4-23-8-3-4-3-8-5,编号0063,与其它93个国内羊种布氏杆菌分离株的型不同,亲缘关系较远,具有独特性。3株菌聚在一起,提示同一地理来源的可能。
[Abstract]:Brucellosis (Brucellosis),) is a zoonotic infectious disease caused by brucellosis (Brucella). In recent years, the epidemic situation of sheep cloth disease in Gansu Province is on the rise, which not only restricts the sustainable development of sheep breeding industry, but also affects the health of the people. In this paper, we investigated the epidemic situation of sheep cloth disease in three areas of Gansu province in 2014. 1. In order to evaluate the diagnostic ability of RBT and I-ELISA in sheep brucellosis without gold standard, RBT and I-ELISA were used to simultaneously detect 176 unimmunized sheep sera collected from a certain place, and a Bayesian model was established. The sensitivity and specificity of the two methods were calculated. The results showed that the sensitivity of RBT and I-ELISA were 76. 5% and 89. 6, and the specificity was 95. 1% and 93. 4% Elisa had better diagnostic ability. 2. 1536 serum samples were collected from 54 farms in Jingyuan County, Kongtong District and Yongchang County. The total positive rate of individuals was 3.58% (55 / 1536) and the total positive rate of population was 25.93% (14 / 54) by I-ELISA. The positive rates of individuals in Jingyuan County, Kongtong District and Yongchang County were 3.35% (18 / 538), 6.02% (30 / 498) and 1.40% (7 / 500) respectively, and the positive rates of population were 22.22% (2 / 9), 33.33% (11 / 33) and 8.30% (1 / 12) respectively. A strain of brucellosis was isolated from Jingyuan County, Kongtong District and Yongchang County in Jingyuan County, Kongtong District and Yongchang County by using brucellosis medium, which was named GS0250,GS2620 and GS2638. in turn. Three strains of Brucella were identified by AMOS-PCR and MLVA-16 by traditional methods. By traditional method, three strains of Brucella sp. 3 were identified as Brucella ovis, and the specific fragments of 731bp were obtained by AMOS-PCR amplification. The MLVA type of the three strains was 1-5-3-13-2-2-2-2-2-4-4-23-8-3-3-8-5, no. 0063, which was different from that of other 93 domestic strains of Brucella. Three strains of bacteria gathered together, indicating the possibility of the same geographical origin.
