AMPK在应激影响卵母细胞发育中的作用

【摘要】:应激可以通过两个方面影响雌性动物的繁殖:(1)干扰排卵;(2)影响植入后的卵母细胞成熟。这个过程是通过HPA轴和HPG轴的交互作用,影响繁殖的顺利进行。繁殖受到能量调控的影响,但调节繁殖的多种生理途径间的关系尚未完全阐明。促性腺激素释放激素(GnRH)神经元通过整合众多生理信号从循环水平调节GnRH的释放,可能会参与调节与生殖有关的能量调控。GnRH神经元可以接收葡萄糖信号,通过AMPK等信号通路进行能量调控。本研究旨在通过证实AMPK参与卵母细胞成熟,AMPK和糖皮质激素有交互影响的作用。最终证明应激条件下,AMPK通路会参与母猪卵母细胞发育。建立卵母细胞体外培养体系,配制母猪卵母细胞成熟培养液。在39℃,CO2浓度5%和饱和湿度的条件下进行培养。使用AMPK激活剂(AICAR)和抑制剂(Compound C)分别处理卵母细胞,统计卵母细胞成熟率;使用糖皮质激素处理卵母细胞,统计卵母细胞成熟率;在添加糖皮质激素处理细胞的同时,分别激活和阻断AMPK信号,检测卵母细胞成熟率的变化。建立小鼠热应激模型,对同期发情并超数排卵处理的小鼠热应激,检测热应激对小鼠繁殖系统及下丘脑AMPK通路、血液指标的影响,使用药物(胆汁酸)对其的治疗作用。进行了两次关于母猪繁殖力的省内调研:第一次以调查问卷形式进行,目的是了解当前省内母猪养殖现状和母猪繁殖力概况,一次以实地调研形式对省内养殖集团下属养殖场进行调研,目的是通过调研,搞清环境中对母猪繁殖力造成影响的应激因素并提出解决建议。
[Abstract]:Stress can affect the reproduction of female animals in two ways: (1) interfere with ovulation and (2) affect oocyte maturation after implantation. This process is affected by the interaction between HPA axis and HPG axis. Reproduction is affected by energy regulation, but the relationship between many physiological pathways regulating reproduction has not been fully clarified. Gonadotropin releasing hormone (GnRH) neurons regulate the release of GnRH from the circulatory level by integrating many physiological signals, which may be involved in regulating the energy regulation associated with reproduction. GnRH neurons can receive glucose signals. The energy is regulated by signal pathways such as AMPK. The aim of this study was to demonstrate that AMPK is involved in oocyte maturation and that AMPK and glucocorticoid play an interactive role in oocyte maturation. Finally, it was proved that AMPK pathway would participate in oocyte development in sows under stress.
