硫磺负离子添加剂对SD大鼠的致畸毒性研究

【摘要】:研究目的:硫磺负离子添加剂是将存在于大自然的多种矿物质用现代高科技纳米技术进行粉碎、筛选、分离、烘干等程序加工,并按合理的科学配比混合而成,可广泛使用于鸡鸭、猪狗、牛羊等家畜饲养及鱼、虾等水产类养殖,也可使用于蔬菜、瓜果类的种植和栽培。本实验主要是通过研究硫磺负离子添加剂对SD大鼠和胎仔致畸敏感期的毒性,为本产品的进一步开发利用提供一定的数据基础。材料和方法:本实验选用健康的SD雌性大鼠,在发情期或发情间期与雄鼠交配,在雌鼠受孕后的5-15d(胎儿发育过程中对受试物最敏感的时期)灌胃10mg/kg、33.3mg/kg、100mg/kg的硫磺负离子添加剂混合液,对照组为1%CMC溶液,至受孕第20d处死动物,检测母体体重、摄食量,观察孕鼠的临床症状及剖检后有无内脏病变的情况;检查胎盘重量、黄体数、着床率、活胎率、胎儿的重量、体长、内脏、骨骼发育情况、雌雄比等,对硫磺负离子添加剂的致畸敏感期生殖毒性进行综合评价。结果:SD雌性大鼠口服硫磺负离子添加剂混合液10mg/kg、33.3mg/kg、 100mg/kg后,对受孕SD雌鼠的摄食量,体重、黄体数、着床率、活胎率;胎仔体重、体长、尾长、肛长,胎仔性别比,胎儿内脏及骨骼检查等各项检测指标同对照组比较差异无统计学意义(P0.05),与SD大鼠致畸敏感期毒性实验基数比较无明显差异,母体无明显毒性反应,胎儿也没出现明显的致畸作用。结论:硫磺负离子添加剂在本实验所用剂量时无毒性和致畸作用,对母鼠的健康及胎鼠的正常发育均无明显毒性和致畸作用。
[Abstract]:Objective: sulfur anion additive is a kind of mineral which exists in nature and is processed by modern high-tech nanotechnology, such as grinding, screening, separating, drying and mixing according to the reasonable scientific proportion. It can be widely used in domestic animals, such as chicken, duck, pig, dog, cattle and sheep, fish, shrimp and other aquaculture, but also can be used in the planting and cultivation of vegetables, melon and fruit. In this experiment, the toxicity of sulfur anion additive to the teratogenic susceptibility of SD rats and foetus was studied, which provided some data basis for the further development and utilization of this product. Materials and methods: healthy female SD rats were used to mate with male rats during estrus or estrus, and 10 mg 路kg ~ (-1) 路kg ~ (-3) mg 路kg ~ (-1) were given to female rats at 5-15 days after conception (the most sensitive period during fetal development). The mixture of sulfur and anion additive in 100mg/kg and 1%CMC solution in control group were used to kill the animals on the 20th day after conception. The maternal body weight and feeding quantity were measured, and the clinical symptoms and visceral lesions of pregnant rats were observed. Placenta weight, luteal body number, implantation rate, live fetus rate, fetal weight, body length, viscera, bone development, ratio of female to male, etc. Results: SD female rats were fed with 10 mg / kg sulfur anion additive (10mg / kg) and 33.3 mg / kg (100mg/kg). The feeding amount, body weight, luteal body number, implantation rate and live birth rate of pregnant SD female rats were measured. There was no significant difference in body weight, body length, tail length, anal length, fetal sex ratio, fetal viscera and bone examination between the two groups (P0.05). There was no significant difference between the experimental cardinal number of teratogenicity and the fetal teratogenicity in SD rats, the maternal toxicity was not obvious and the fetus had no obvious teratogenicity. Conclusion: the sulfur anion additive has no toxicity and teratogenicity at the dose of this experiment, but has no obvious toxicity and teratogenicity on the health of the mother and the normal development of the fetal mouse.
