维氏气单胞菌JLL-1株主要生物学特性及灭活疫苗免疫效果评价

【摘要】:维氏气单胞菌(Aeromonas veronii AV)是一种人兽鱼共患病病原菌,其危害程度已经引起有关专家学者的重视,部分国家已经将该菌列为食品安全的检疫项目之一,但是有关维氏气单胞菌的研究进展较为缓慢。因此本实验以分离自吉林某鲤鱼场的JLL-1菌株为实验材料,对其主要生物学特性进行研究,为维氏气单胞菌的鉴定提供进一步的理论指导。由于灭活疫苗,其制备方便,免疫效果好,容易受到广大养殖户的青睐,因此本实验对JLL-1菌株进行灭活疫苗免疫效果的观察。本实验首先对维氏气单胞菌JLL-1菌株进行主要生物学特性研究:通过革兰氏染色镜检观察该菌株为革兰氏阴性短杆菌;在液体LB培养基中可出现二次生长的现象;可在EMB培养基、BS培养基和SS培养基等多种培养基中生长良好;其耐受性较强,可在温度25℃~42℃、pH值6~10、NaCL含量0%~4%中生长;药物敏感实验结果分析得出,JLL-1菌株对16种药物敏感,约占检测药物的69.57%,而处于中介及耐药药物分别占测定药物的17.39%和13.04%;该菌的半数致死量为3.98×104CFU/mL,可引起鱼鳍、鳃出血,腹腔有大量黄色或带血腹水,肠管充血,呈急性死亡等症状。对灭活疫苗组及PBS组在免疫前0周及免疫后第1周、第2周、第3周、第4周每组随机取10尾鱼尾静脉采血,部分抗凝血用于白细胞吞噬实验,其余则制备血清,用于测定各免疫指标。结果显示,JLL-1灭活疫苗可有效增强锦鲤的免疫力:酸性磷酸酶(ACP)在免疫1周后呈显著上升趋势,在第二周达到峰值104.6金氏单位/100mL后有所下降,但仍高于对照组;碱性磷酸酶(AKP)则呈先下降后上升趋势,但仍达不到免疫前的水平;超氧化物歧化酶(SOD)呈显著上升趋势,免疫2周后达到最大值224U/mL,之后下降,但在第4周仍高于对照组;白细胞吞噬指数(PI)也呈上升趋势,在第3周达到最大值2.9;白细胞吞噬百分比(PP)也同样在第3周达到最大值36.67%,其变化趋势与白细胞吞噬指数(PI)一致;血清的杀菌活性则在免疫第1周到第4周都维持在较高水平,其细菌的存活指数分别为59.42、36.57、8.23和46.49;免疫组抗体效价从第0周的1:40开始上升,第1周为1:160,在第2周达到1:320,第3周达到最大抗体效价1:1280,第4周下降到1:640,而对照组抗体效价始终维持在1:40;免疫四周后进行攻毒试验,经14d观察计算出相对免疫保护率为88%,有较好的保护性。
[Abstract]:Aeromonas Vickers (Aeromonas veronii AV) is a zoonotic pathogen of human orcs, which has attracted the attention of relevant experts and scholars, and has been listed as one of the quarantine items in food safety in some countries. However, the progress of Aeromonas Vickers is relatively slow. Therefore, the main biological characteristics of JLL-1 strains isolated from a carp farm in Jilin Province were studied in order to provide further theoretical guidance for the identification of Aeromonas Vickers.
