滩羊同期发情及精液低温保存技术研究

【摘要】:现代肉羊生产体系就是要以适繁母羊为基础,充分发挥其繁殖潜力,以先进的科学技术手段为依托,提高其繁殖力。而宁夏滩羊繁殖还停留在传统的自然交配,母羊繁殖力低,国内外重要的先进技术手段的价值没有得到应有的发挥和体现,同期发情、人工授精等繁殖技术没有得以广泛推广。本试验针对宁夏滩羊养殖现状,对宁夏滩羊进行同期发情技术和精液低温保存技术的应用性研究。通过不同的同期发情处理方案,研究出效果最佳的同期发情方法;通过对不同配方低温保存稀释液对公羊精液保存效果的研究,筛选出低温条件下保存滩羊精液的最佳稀释剂配方;通过同期发情技术、发情鉴定、人工采精、精液稀释保存技术、人工授精技术以及妊娠诊断技术等一系列繁殖技术的综合应用,探索提高滩羊繁殖力的技术方法。本试验的主要研究结果如下:1.使用“阴道栓+FSH+PG”处理的试验组羊的同期发情率和配种受胎率均高于“PG+FSH”处理的试验组(P0.05),注射FSH 50 IU+PG 0.1 mg 96 h后放置NRID处理13 d,分别在-24 h,-12 h注射FSH 50 IU,0 h去栓并注射PG 0.1 mg的同期发情方案效果最佳,使母羊同期发情率达到100%,极显著高于其他试验组(P0.01);母羊情期受胎率达到75.3%,显著高于其他各组(P0.05),且发情时间相对集中于24~48 h。2.试验所选用的6种稀释液配方以5号保存效果最好,保存时间和有效存活时间及生存指数显著高于其他配方(P0.05),6号次之。用5号稀释液保存精液,使滩羊精液最长存活162 h,精子有效存活时间长达87±5 h,生存指数89.64±5.1,24 h内配种受胎率50%以上。3.精液稀释液中添加海藻糖能够显著提高精子活率、精子运动性能以及精子质膜完整性(P0.05)。在本试验所设计的浓度范围内,随海藻糖浓度增加,精子活率并无显著变化(P0.05),精子质膜完整率和精子运动性能均呈成上升趋势,但这种变化趋势不显著(P0.05)。相同保存时间下,浓度为0.03 g/mL时质膜完整率比添加其他浓度时高,差异不显著(P0.05),精子运动速度也高于其他添添加浓度,差异不显著(P0.05)。
[Abstract]:The production system of modern meat sheep is to make full use of its breeding potential and to improve its reproduction ability based on advanced scientific and technological means. However, the breeding of Ningxia Tan sheep still stays in the traditional natural mating, the fecundity of the ewes is low, the value of the important advanced technical means at home and abroad has not been given full play and embodiment, and the breeding techniques such as simultaneous estrus and artificial insemination have not been widely popularized. Based on the present situation of Ningxia Tan sheep breeding, the application of synchronized estrus and semen cryopreservation technique was studied. Through different estrus synchronization treatment schemes, the best estrus synchronization method was developed. The best diluent formula for the preservation of Tan sheep semen at low temperature was obtained by studying the effects of different low temperature preservation diluents on the preservation of ram semen. Through the comprehensive application of a series of breeding techniques, such as simultaneous estrus technology, estrus identification, artificial semen collection, semen dilution and preservation technology, artificial insemination technology and pregnancy diagnosis technology, this paper explores the technical methods to improve the fecundity of Tan sheep. The main results of this study are as follows: 1. The rate of synchronous estrus and fecundity of sheep treated with "vaginal suppository FSH PG" was higher than that of "PG FSH" treatment group (P0.05). The treatment group was treated with NRID for 13 days after injecting FSH 50 IU PG 0.1 mg for 96 h, and was treated with NRID for -24 h, respectively. The estrus synchronization rate of estrus was 100, which was significantly higher than that of other experimental groups (P0.01). The conception rate of estrus in estrus was 75.3, which was significantly higher than that in other groups (P0.05), and the estrus time was relatively concentrated at 244h.2. The six diluent formulations selected in the experiment had the best preservation effect, and the preservation time, effective survival time and survival index were significantly higher than those of other formulations (P0.05), followed by No. 6. The semen of Tan Sheep was preserved with dilution No. 5 for the longest time of 162 hours, the effective survival time of sperm was 87 卤5 h, and the survival index was 89.64 卤5.1 鈩,
