郁金散对大肠湿热证模型大鼠血清及肠道组织胃肠激素的影响

【摘要】:探讨郁金散对大肠湿热证模型大鼠血清和肠道组织胃肠激素的影响。Wistar大鼠60只,体重180~220g,随机分为正常对照组、大肠湿热证模型组、自愈组、郁金散高、中、低剂量治疗组,每组10只。通过自由饮用蜂蜜水;前10d禁食与饲喂充足饲料并灌胃猪油隔日交替进行;之后5d每天灌白酒,并置于高温高湿环境(8h·d-1);然后腹腔注射大肠杆菌,24h后再注射一次建立大肠湿热证大鼠模型。造模成功后,分别给予高(10.4g·kg~(-1))、中(5.2g·kg~(-1))、低(2.6g·kg~(-1))剂量郁金散进行治疗。整个试验过程中观察大鼠的临床症状和体征,记录体重变化,检测血清和回肠、结肠组织中的胃肠激素含量。结果显示:模型大鼠出现便溏、粪便色黄恶臭、肛门红肿、发热等症状和体征。正常组体重逐渐上升,模型大鼠造模期间体重呈现先波状上升、再逐渐下降的趋势,郁金散治疗后体重呈逐渐上升的趋势,其中以高剂量组回升最明显,但各组均无显著性差异(P0.05)。与正常对照组相比,模型组和自愈组大鼠血清、回肠及结肠组织中胃动素(MTL)、胃泌素(GAS)、P物质(SP)含量极显著或显著升高(P0.01或P0.05),血管活性肠肽(VIP)无显著变化,生长抑素(SS)含量极显著降低(P0.01);与自愈组相比,各剂量郁金散治疗后,MTL、GAS、SP、VIP和SS都有不同程度的回调,以高剂量组回调最为明显,其中高剂量组大鼠血清、回肠、结肠中MTL、GAS和SP极显著或显著降低(P0.01或P0.05),SS极显著或显著升高(P0.01或P0.05)。大肠湿热证模型大鼠机体出现了胃肠激素紊乱,表现为MTL、GAS、SP和VIP含量升高,而SS含量降低,郁金散能改善大肠湿热证模型大鼠机体胃肠激素异常水平,且高剂量的治疗效果最好。
[Abstract]:To explore the effect of Yujin Powder on gastrointestinal hormones in serum and intestinal tissue of rats with large intestine damp-heat syndrome. Sixty Wistar rats, weighing 180g / 220 g, were randomly divided into three groups: normal control group, model group of large intestine damp-heat syndrome, self-healing group, high, middle and low dose treatment group. 10 rats in each group. After drinking honey water freely, fasting for the first 10 days, feeding enough feed and gavage lard alternately, after 5 days, liquor was infused daily and placed in high temperature and high humidity environment (8h 路d ~ (-1). Then the coliform was injected intraperitoneally, and the rat model of large intestine damp-heat syndrome was established again after 24 hours. After the model was successfully established, the high (10.4 g kg~ (-1),) (5.2 g kg~ (-1),) low dose (2.6 g kg~ (-1) of Yujin Powder) was given respectively. The clinical symptoms and signs of the rats were observed, the changes of body weight were recorded, and the contents of gastrointestinal hormones in serum, ileum and colon were detected. The results showed that the model rats had symptoms and signs, such as loose stools, yellow stench of faeces, red and swollen anus, fever and so on. The body weight of the normal group gradually increased, and the weight of the model rats increased at first, then decreased gradually, and the weight increased gradually after the treatment of Yujin Powder, especially in the high dose group. But there was no significant difference in each group (P0.05). Compared with normal control group, the content of motilin (MTL), gastrin (GAS), P (SP) in serum, ileum and colon of model group and self-healing group was significantly higher than that of normal control group (P0.01 or P0.05). There was no significant change in vasoactive intestinal peptide (VIP), but the content of somatostatin (SS) was significantly decreased (P0.01). Compared with the self-healing group, MTL,GAS,SP,VIP and SS had different degrees of callback, especially in the high-dose group, in which the MTL, in serum, ileum and colon of rats in the high-dose group was more obvious than that in the self-healing group. GAS and SP were significantly decreased (P0.01 or P0.05), SS significantly or significantly increased (P0.01 or P0.05). Gastrointestinal hormone disorder appeared in rats with large intestine damp-heat syndrome, which showed that the contents of MTL,GAS,SP and VIP increased, while the content of SS decreased. Yujin Powder could improve the abnormal level of gastrointestinal hormones in rats with large intestine damp-heat syndrome. And the high dose of the treatment is the best.
