肉鸡腹水综合征(AS)中MIF与AMPK表达关系的研究

【摘要】:[目的]肉鸡腹水综合征(AS)严重影响养鸡业经济效益和肌肉品质。以往主要集中于研究缺氧引起的肺动脉高压环节,但是不能全面揭示该病的发病机制。从病理解剖和病理生理看,该病的显著特征是右心肥大,心脏是肉鸡腹水综合征的主要靶器官。本文认为肉鸡腹水综合征本质上属代谢性疾病,慢性心衰导致肉鸡腹水综合征。AMPK是心肌能量代谢平衡调节器,巨噬细胞移动抑制因子(MIF)是影响AMPK活性的重要细胞因子。因此,本文从肉鸡心脏代谢角度入手研究肉鸡腹水综合征。[方法]本研究以临床上的腹水综合征肉鸡为研究对象,采用免疫印迹方法检测MIF、AMPK、p-AMPK在临床健康肉鸡与腹水综合征肉鸡心脏组织的表达水平,以探究肉鸡腹水综合征中MIF与AMPK的表达关系。[结果]1采用免疫印迹方法对MIF在临床健康肉鸡心脏、肝脏、脾脏、肺脏、肾脏的表达情况进行分析,结果显示:MIF在各组织中均有表达。2采用免疫印迹方法对MIF在临床健康肉鸡与腹水综合征肉鸡中心脏、肝脏、脾脏、肺脏、肾脏的表达情况进行分析,结果显示:MIF在腹水综合征肉鸡的心脏、肝脏表达显著增加,在肾脏的表达显著下降,在脾脏和肺脏的表达增加,但差异不显著。3采用免疫印迹方法对AMPK、p-AMPK在腹水综合征肉鸡心脏中的表达情况进行分析,结果显示:腹水综合征肉鸡的MPK磷酸水平显著升高。[结论]肉鸡腹水综合征发生时,心肌MIF显著增加,AMPK显著活化,减少心肌损伤,为AMPK信号通路调节AS的心肌能量代谢机制提供了新思路。
[Abstract]:[objective] (AS) of broiler ascites syndrome seriously affected the economic benefit and muscle quality of chicken industry. In the past, it was mainly focused on the study of hypoxic pulmonary hypertension, but it can not fully reveal the pathogenesis of the disease. According to pathological anatomy and pathophysiology, the disease is characterized by the fat right heart and the heart is the main target organ of ascites syndrome in broilers. It is considered that ascites syndrome in broilers is essentially a metabolic disease, chronic heart failure leads to ascites syndrome in broilers, AMPK is a balance regulator of myocardial energy metabolism, and macrophage migration inhibitor (MIF) is an important cytokine affecting the activity of AMPK. Therefore, this paper studies ascites syndrome in broiler from the angle of heart metabolism. [methods] the expression of MIF,AMPK,p-AMPK in the heart of clinical broilers with ascites syndrome was detected by Western blot. To explore the expression of MIF and AMPK in broiler ascites syndrome. [results] 1the expression of MIF in heart, liver, spleen, lung and kidney of healthy broilers was analyzed by Western blot. The results showed that MIF was expressed in all tissues. 2 the expression of MIF in heart, liver, spleen, lung and kidney of clinical healthy broilers and broilers with ascites syndrome was analyzed by Western blot. The results showed that the expression of MIF in the heart and liver of broilers with ascites syndrome was significantly increased, the expression in kidney was decreased significantly, the expression in spleen and lung was increased, but the difference was not significant. 3 Immunoblot method was used to detect AMPK,. The expression of p-AMPK in the heart of broilers with ascites syndrome was analyzed. The results showed that the level of MPK phosphoric acid in broilers with ascites syndrome was significantly increased. [conclusion] during the occurrence of ascites syndrome in broilers, myocardial MIF was significantly increased, AMPK was activated significantly and myocardial injury was reduced, which provided a new idea for the regulation of myocardial energy metabolism mechanism of AS by AMPK signaling pathway.
