鸡免疫器官及血液中淋巴细胞变化规律的研究

【摘要】:淋巴细胞主要包括T淋巴细胞和B淋巴细胞,分别参与细胞免疫和体液免疫。本研究应用流式细胞术,以SPF鸡为研究对象,分别在1、4、7、14、21、35、56和90日龄采取鸡的法氏囊、胸腺、脾脏以及血液,检测其在正常发育过程中T、B淋巴细胞数量的变化情况。通过用流式细胞仪检测B淋巴细胞中Bu-1b以及IgA+、IgG+和IgM+细胞数量的变化情况,结果发现:1日龄时Bu-1b细胞数量较少,4日龄时免疫器官及外周血液中的Bu-1细胞数量迅速增多,7日龄时,法氏囊中的Bu-1b细胞达到增长的高峰,说明在4至7日龄时体液免疫功能增强;在脾脏、胸腺、法氏囊以及血液中,B淋巴细胞中的免疫球蛋白主要以IgM+细胞数量为主,Ig G+细胞数量最少;此外研究还发现,免疫器官及外周血液中的B淋巴细胞的数量在35日龄基本达到稳定。通过对T细胞数量的变化进行检测,结果显示:在1至4日龄时CD3+细胞数量迅速增多,7日龄时法氏囊中CD3+细胞数量减少,到35日龄以后CD3+细胞数量变化幅度不大,基本趋于稳定;14日龄时,血液和胸腺中的CD4+细胞含量多于CD8+细胞含量,而在21日龄时法氏囊中的CD4+细胞含量多于CD8+细胞的含量。在21日龄时CD4+细胞数量趋于稳定。本研究利用流式细胞术研究SPF鸡在正常生理情况下,免疫器官和血液中T、B淋巴细胞及其亚群中细胞数量的变化规律,从而了解到鸡出壳后的免疫状态,这将为适时免疫和疾病监测提供了重要的理论依据。
[Abstract]:Lymphocytes mainly include T lymphocytes and B lymphocytes, which participate in cellular immunity and humoral immunity respectively. In this study, flow cytometry was used to investigate the changes of T B lymphocytes in the bursa of Fabricius, thymus, spleen and blood of SPF chickens during normal development. The changes of Bu-1b, IgA, IgG and IgM cells in B lymphocytes were detected by flow cytometry. The results showed that the number of Bu-1b cells was less at 1 day old. The number of Bu-1 cells in immune organs and peripheral blood increased rapidly at the age of 4 days, and the Bu-1b cells in the bursa of Fabricius reached the peak at the age of 7 days, indicating the enhancement of humoral immune function at the age of 4 to 7 days. In spleen, thymus, bursa of Fabricius and blood, the number of immunoglobulin in B lymphocytes was mainly IgM cells, mainly, Ig G cells. In addition, the number of B lymphocytes in immune organs and peripheral blood was stable at 35 days old. The results showed that the number of CD3 cells increased rapidly at the age of 1 to 4 days, the number of CD3 cells in the bursa of Fabricius decreased at the age of 7 days, and the number of CD3 cells did not change significantly after 35 days of age. It tends to be stable; At the age of 14 days, the content of CD4 cells in blood and thymus was higher than that of CD8 cells, and the content of CD4 cells in bursa of Fabricius was higher than that of CD8 cells in the bursa of Fabricius at 21 days of age. At 21 days of age, the number of CD4 cells tended to stabilize. In this study, flow cytometry was used to study the changes of the number of TG-B lymphocytes and their subsets in the immune organs and blood of SPF chickens under normal physiological conditions. This will provide an important theoretical basis for timely immunization and disease surveillance.
