褪黑激素对内蒙古绒山羊产绒性能及皮肤毛囊中RORa表达的影响

【摘要】:内蒙古绒山羊是我国绒山羊珍贵品种之一,其中绒纤维经济价值较高,素有“纤维宝石”之称。通过前人研究,发现褪黑激素能够促进绒毛生长,但其信号通路及调控机理尚不清楚。因此,本研究以内蒙古绒山羊为研究对象,在耳后皮下埋植褪黑激素,对绒毛生长特性与毛品质进行检测,并利用Western Blot和免疫荧光技术探讨褪黑激素对核受体RORa在内蒙古绒山羊2、9、12月份皮肤毛囊中蛋白水平的表达及分布的影响。研究得到结果如下:(1)绒毛生长特性与毛品质检测结果表明:试验组与对照组的羊绒细度在9月份和12月份差异显著(P0.05),其他各月份差异均不显著(P0.05);试验组与对照组羊绒长度在9、10、11、1、2月份和3月份差异均显著(P0.05),8、12和4月份差异均不显著(P0.05);结果表明埋植褪黑素有促进绒毛变长的趋势;试验组与对照组的产绒量差异不显著(P0.05),但产绒量有增加的趋势。结果表明褪黑激素对绒毛的长度影响较大、对绒毛细度和产绒量影响不大,但是产绒量有提高的趋势。(2) Western Blot结果表明:RORa蛋白在2、9、12月份的对照组和试验组的皮肤毛囊中均表达,且2月份表达量最高,9月份下降,12月份最低;2月份试验组RORa蛋白表达量显著高于对照组(P0.05);在9、12月份试验组与对照组组RORa蛋白表达量均差异不显著(P0.05),结果表明褪黑激素上调了2月份RORa蛋白的表达。(3)免疫荧光染色结果表明:对照组中2、9、12月份RORa蛋白只在毛囊外根鞘表达,而试验组中3个月份RORa蛋白在毛囊外根鞘和毛乳头中均有表达。本试验研究埋植和不埋植褪黑激素对绒毛产量与绒毛品质的影响,利用Western Blot技术和免疫荧光检测技术,分析埋植褪黑激素对绒山羊皮肤毛囊中RORa表达的影响,结果表明褪黑激素促进绒毛生长是与毛囊外根鞘和毛乳头中的RORa结合,进而参与调节毛囊周期性生长。
[Abstract]:Inner Mongolia cashmere goat is one of the precious breeds of cashmere goat in China, in which cashmere fiber is of high economic value and is known as "fiber gem". By previous studies, it was found that melatonin can promote villus growth, but its signaling pathway and regulation mechanism are not clear. Therefore, in this study, Inner Mongolia cashmere goats were implanted subcutaneously with melatonin in the back of ear to detect the growth characteristics and hair quality of hair. The effects of melatonin on the expression and distribution of nuclear receptor RORa in the hair follicles of Inner Mongolia Cashmere Goats were studied by Western Blot and immunofluorescence techniques. The results were as follows: (1) the results showed that the cashmere fineness of the experimental group and the control group were significantly different in September and December (P0.05), but there was no significant difference in the other months (P0.05). The cashmere lengths of the experimental group and the control group were significantly different in 9, 10, 11, 1, 2 and 3 months (P0.05), but there was no significant difference in 8,12 and April (P0.05), the results showed that melatonin implantation had a tendency to promote villus lengthening. There was no significant difference between the treatment group and the control group (P0.05), but there was an increasing trend of the cashmere yield. The results showed that melatonin had a great effect on the length of villus, but had little effect on the fineness and yield of villus. (2) Western Blot results showed that RORa protein was 2,9, The expression in the hair follicles of the control group and the experimental group in December was the highest in February, decreased in September, and the lowest in December. In February, the expression of RORa protein in experimental group was significantly higher than that in control group (P0.05). There was no significant difference in the expression of RORa protein between the experimental group and the control group in September and December (P0.05), the results showed that melatonin upregulated the expression of RORa protein in February. (3) the immunofluorescence staining results showed that in the control group, the expression of RORa protein was significantly increased. The RORa protein was only expressed in the outer root sheath of the hair follicle in December, while the RORa protein was expressed in the outer root sheath and papilla of the hair follicle in the experiment group for three months. In this study, the effects of melatonin on the yield and quality of cashmere were studied. The effects of melatonin on the expression of RORa in the hair follicles of cashmere goats were analyzed by Western Blot technique and immunofluorescence assay, and the effect of melatonin on the expression of melatonin in the hair follicles of cashmere goats was studied. The results showed that melatonin promoted villous growth by binding to RORa in the outer root sheath and papilla of hair follicles, and then involved in regulating the periodic growth of hair follicles.
