奶山羊FABP3和FABP4基因的多态性与乳脂肪酸成分的关联分析

【摘要】:脂肪酸结合蛋白(Fatty acid binding protein,FABP)是与胞内脂质相结合的一类蛋白,其对长链脂肪酸具有高度的亲和力。长链脂肪酸从血液进入细胞后,需与FABP结合,运输到特定的靶向细胞器。因此在动物体内,FABP对脂肪酸的摄取、胞内转运、脂化、氧化以及合成均有重要作用。在奶山羊的乳腺组织中,FABP3和FABP4基因的表达量丰富,故FABP3和FABP4基因遗传信息的变化,将直接或间接地调控羊奶成分与脂肪酸的组成。本试验以西农萨能奶山羊为研究对象,分析FABP3和FABP4基因的遗传多态性,采集不同月份泌乳羊奶样(泌乳30 d,60 d,90 d,120 d)并分析其主要乳成分和脂肪酸组成,筛选出显著性影响奶山羊泌乳性状的SNP。主要研究结果如下:(1)采用DNA池测序和PCR-RFLP等方法研究了FABP3和FABP4基因的遗传变异,检测到4个新的多态位点,分别位于FABP3基因第一外显子的SNP G-986-A,FABP3基因第三内含子的SNP C-3064-A,FABP4基因第二外显子的SNP G-3632-A,FABP4基因第二内含子的SNP A-3716-G。其中SNP G-986-A为错义突变(Cys20Tyr),SNP G-3632-A为同义突变(Arg60Arg)。(2)通过对FABP3和FABP4基因多态性的分析,发现4个多态位点的基因(型)频率,其多态信息含量均在30%以上,并符合哈代-温伯格平衡定律。其中,FABP4基因的SNP G-3632-A和SNP A-3716-G高度连锁,FABP4基因的单倍型-GA-出现的频率最高,达59.0%。(3)羊奶中测得37种脂肪酸,其中油酸含量最高,羊奶中饱和脂肪酸与不饱和脂肪酸含量的比例为2:1。随着泌乳时间的延长,羊奶中饱和脂肪酸的含量增加,然而不饱和脂肪酸的含量降低。(4)对FABP3和FABP4基因的多态位点与羊奶成分进行关联分析,发现FABP3基因的SNP G-986-A位点与奶山羊的乳脂、总脂固形物和长链脂肪酸的含量以及脂肪酸的不饱和度显著相关;FABP3基因的SNP C-3064-A位点与奶山羊的长链脂肪酸含量以及脂肪酸的不饱和度显著相关;FABP4基因的SNP G-3632-A与奶山羊的乳脂、乳蛋白、总脂固形物、非脂固形物和长链脂肪酸含量以及脂肪酸的不饱和度显著相关;FABP4基因的SNP A-3716-G与奶山羊的长链脂肪酸含量以及脂肪酸的不饱和度显著相关。
[Abstract]:Fatty acid binding protein (Fatty acid binding protein,FABP (fatty acid binding protein) is a kind of protein that binds to intracellular lipid and has high affinity to long chain fatty acid. After entering cells from blood, long chain fatty acids bind to FABP and transport to specific targeted organelles. Therefore, FABP plays an important role in fatty acid uptake, intracellular transport, lipid formation, oxidation and synthesis in animals. The expression levels of FABP3 and FABP4 genes are abundant in breast tissues of dairy goats. Therefore, the changes of genetic information of FABP3 and FABP4 genes will directly or indirectly regulate the composition of goat milk components and fatty acids. The genetic polymorphisms of FABP3 and FABP4 genes were analyzed in Xinong Saanen dairy goats in this experiment. Milk samples (30 d, 60 d, 90 d, 120 d) of lactating sheep in different months were collected and their main milk composition and fatty acid composition were analyzed. Screening of SNP. with significant effect on lactation of Dairy Goat The main results are as follows: (1) the genetic variation of FABP3 and FABP4 genes was studied by DNA pool sequencing and PCR-RFLP. In the third intron of the FABP3 gene, the SNP Cx3064A, the second exon of the FABP4 gene, and the second intron of the FABP4 gene, the SNP GX3632A, and the second intron of the FABP4 gene, SNP Ax3716G. Among them, SNP GX986A is missense mutation (Cys20Tyr), SNP GX3632A is synonymous mutation (Arg60Arg). (2) by analyzing the polymorphism of FABP3 and FABP4 genes, the gene frequencies of four polymorphic loci were found, and the gene frequencies of the four polymorphic loci were detected by the analysis of the polymorphism of the FABP3 and FABP4 genes. The polymorphic information content is more than 30% and accords with Hardy-Weinberg equilibrium law. The haplotype-GA- of FABP4 gene had the highest frequency of 59.0%. (3) 37 kinds of fatty acids were detected in goat milk, among which oleic acid content was the highest, and the frequency of haplotype-GA- of FABP4 gene was the highest in goat milk, the highest in which oleic acid content was found in sheep milk, and the frequency of haplotype-GA- in FABP4 gene was the highest (59.0%). The ratio of saturated fatty acid to unsaturated fatty acid in goat milk is 2. With the prolongation of lactation time, the content of saturated fatty acids in goat milk increased, but the content of unsaturated fatty acids decreased. (4) the polymorphism sites of FABP3 and FABP4 genes were associated with the composition of goat milk. It was found that there was a significant correlation between the SNP G / 986 A site of FABP3 gene and milk fat, total fat solids, long chain fatty acids and unsaturation of fatty acids in dairy goats. There was a significant correlation between the SNP C _ (3 064) A site of FABP3 gene and the content of long chain fatty acids and the unsaturation of fatty acids in dairy goats. The SNP of FABP4 gene was significantly correlated with milk fat, milk protein, total fat solids, non-fat solids, long-chain fatty acids and unsaturation of fatty acids in dairy goats. The SNP of FABP4 gene was significantly correlated with the content of long chain fatty acids and the unsaturation of fatty acids in dairy goats.
