AMPK信号通路与高能低蛋白日粮致蛋鸡FLHS的关系研究

【摘要】:蛋鸡脂肪肝出血综合征(FLHS)是蛋鸡体内脂肪代谢紊乱引起的以肝脏发生脂肪变性、出血为特征的一种营养代谢病,是高产蛋鸡常见多发病。高能低蛋白日粮是我国目前生产上诱发蛋鸡脂肪肝出血综合征的主要原因之一。近年来研究发现一磷酸腺苷(AMP)激活的蛋白激酶(AMPK)在能量代谢中起着重要作用(特别是在脂质代谢中)。本试验旨在研究AMPK信号通路与高能低蛋白日粮致蛋鸡脂肪肝出血综合征之间的关系,选用200羽海兰褐蛋鸡随机分成两组,对照组和试验组,每组5个重复,每个重复20羽,对照组饲喂基础日粮代谢能(2678.99 kcal/kg,粗蛋白15.86%),试验组饲喂高能低蛋白日粮(代谢能3100.00 kcal/kg,粗蛋白12.00%),试验期为100d,分别于试验的第20d、40d、60d、80d和100d每组随机选取20羽蛋鸡进行采样,采集血液、肝脏和下丘脑组织,制作肝脏组织的病理切片,检测血清和肝脏组织中的TG、TC、LDL、HDL和NEFA的含量,检测肝脏和下丘脑中AMPK、ACC、LKB1、HMGR、FAS、GPAT等相关因子的mRNA的表达量。结果如下:1.试验组大量肝细胞肿胀,细胞膜模糊;胞浆内可见数量不等、大小不一的脂肪空泡;细胞核被推向一侧或消失。2.与对照组比较,试验组血清和肝脏组织匀浆中TG、TC、LDL和NEFA的含量显著上升,HDL的含量显著下降(P0.05)。3.与对照组比较,试验组肝脏和下丘脑中的ACC、HMGR、FAS、GPAT、HNF4α和PPARα的mRNA的表达量显著上升(P0.05),AMPKα1、LKB1和CPT1的mRNA的表达量显著下降(P0.05)。结论:高能低蛋白日粮诱导的蛋鸡脂肪肝出血综合征能够引起肝脏的脂肪病变,引起肝脏和下丘脑中AMPK的mRNA的表达量下降,造成脂肪酸和胆固醇的合成增多,而脂肪酸和胆固醇的分解减少,进而加剧了蛋鸡肝脏中的胆固醇和脂质沉积。
[Abstract]:Fatty liver hemorrhage syndrome (FLHS) is a nutritional metabolic disease characterized by fatty degeneration and hemorrhage of liver caused by fat metabolism disorder in laying hens. It is a common and frequent disease in high-yielding laying hens. High energy and low egg white diet is one of the main causes of fatty liver bleeding syndrome in laying hens. In recent years, it has been found that adenosine monophosphate (AMP)-activated protein kinase (AMPK) plays an important role in energy metabolism (especially in lipid metabolism). The purpose of this study was to study the relationship between AMPK signaling pathway and fatty liver hemorrhage syndrome in laying hens induced by high energy and low egg diet. The two groups were randomly divided into two groups: control group and experimental group. The control group was fed with basal diet (2678.99 kcal/kg, crude protein 15.86%), the experimental group was fed with high energy low egg white diet (metabolic energy 3100.00 kcal/kg, crude protein 12.00%), the experimental period was 100 days, respectively, on the 20th day of the experiment, the experimental group was fed with high energy low egg white diet (3100.00 kcal/kg, crude protein 12.00%). At 40 days, 60 days, 80 days and 100 days, 20 layers of hens in each group were randomly selected to sample the blood, liver and hypothalamus tissue, make pathological sections of liver tissue, and detect the contents of TG,TC,LDL,HDL and NEFA in serum and liver tissue. The expression of mRNA of AMPK,ACC,LKB1,HMGR,FAS,GPAT and other related factors in liver and hypothalamus were detected. The results are as follows: 1. In the experimental group, a large number of hepatocytes were swollen and the cell membrane was blurred; fat vacuoles were seen in the cytoplasm in varying numbers and sizes; the nucleus was pushed to one side or disappeared. Compared with the control group, the contents of TG,TC,LDL and NEFA in serum and liver homogenate of the experimental group increased significantly, while the content of HDL decreased significantly (P0.05). Compared with the control group, the mRNA expression of ACC,HMGR,FAS,GPAT, HNF4伪 and PPAR 伪 in the liver and hypothalamus of the experimental group increased significantly (P0.05), while the mRNA expression of AMPK伪 1, LKB1 and CPT1 decreased significantly (P0.05). Conclusion: fatty liver hemorrhage syndrome induced by high-energy and low-egg diet can cause fatty liver fat lesion, decrease the expression of AMPK mRNA in liver and hypothalamus, and increase the synthesis of fatty acids and cholesterol. The degradation of fatty acids and cholesterol increases cholesterol and lipid deposition in the liver of laying hens.
