黔北麻羊繁殖相关基因mRNA表达水平与多态性分析

【摘要】:本研究以黔北麻羊为主要研究对象,分析FSHβ、FSHR、Gn RH和Gn RHR基因多态性与繁殖力的关系,Gn RHR和FSHR在发情间期和发情期的表达规律,Gn RHR基因CDS区功能及结构。本试验旨在研究黔北麻羊繁殖相关基因多态与表达在低产和高产山羊的差异,为从分子遗传方法上提高山羊繁殖力提供科学依据。研究结果如下:1、FSHβ和Gn RH基因中没有检测出多态位点,均不具有多态性。2、FSHR基因外显子1的C126T和第10外显子的C1246A和C1412A发现3个SNP位点。遗传变异与产羔数关联分析,C1246A位点,黔北麻羊个体产羔数存在差异,C1412A位点黔北麻羊和贵州黑山羊个体产羔数存在差异;在Gn RHR基因第1外显子发现G121A突变。分析发现,在黔北麻羊和贵州黑山羊两个群体个体的产羔数存在明显差异。3、在发情间期,黔北麻羊BB基因型个体下丘脑和子宫FSHR的表达量高于AA基因型个体(P㩳0.05);黔北麻羊AA基因型垂体、输卵管、卵巢和子宫Gn RHR的表达量高于GG基因型黔北麻羊(P㩳0.05)。在发情期,黔北麻羊BB基因型个体卵巢和子宫FSHR的表达量高于AA基因型个体(P㩳0.05);黔北麻羊AA基因型垂体、输卵管、卵巢和子宫Gn RHR的表达量高于GG基因型黔北麻羊(P㩳0.05)。4、在黔北麻羊垂体中成功克隆Gn RHR基因CDS区984bp,可编码328个氨基酸。
[Abstract]:In this study, Guizhou Ma sheep was used as the main research object to analyze the relationship between FSH 尾, FSHR,Gn RH and Gn RHR gene polymorphism and fecundity. The expression of, Gn RHR and FSHR during estrus and estrus was analyzed. The function and structure of, Gn RHR gene CDS region. The purpose of this study was to study the difference between the polymorphism and expression of reproductive related genes in low yield and high yield goats, and to provide scientific basis for improving the fecundity of goats by molecular genetic method. The results were as follows: 1. No polymorphism was detected in Gn RH 尾 and gene. 2, three SNP loci were found in C126T in Exon 1 and C1246A and C1412A in Exon 10. The association analysis between genetic variation and lambing number showed that there were differences in C1246A locus and individual lambing number between Guizhou and Guizhou black sheep, and G121A mutation was found in exon1 of Gn RHR gene in Guizhou black sheep and Guizhou black sheep, and there were differences in individual lambing number between Guizhou black sheep and Guizhou black sheep at C1246A locus and individual lambing number in Qianbei Ma sheep and Guizhou black sheep at C1412A locus. It was found that there was a significant difference in the number of lambs between Guizhou black sheep and Guizhou black sheep. 3, in the estrus period, The expression of FSHR in Hypothalamus and uterus of individuals with BB genotype was higher than that of individuals with AA genotype (P 鈮,
