硫酸化银耳多糖和党参多糖对鸡T淋巴细胞增殖及白细胞介素-2mRNA表达水平的影响

【摘要】:为研究硫酸化银耳多糖(sTPS_(70c))和硫酸化党参多糖(sCPPS_(50c))增强免疫作用机理,本试验以未修饰银耳多糖(TPStp)为对照,采用噻唑蓝(MTT)法和实时荧光定量PCR法测定了sTPS_(70c)和sCPPS_(50c)对鸡T淋巴细胞增殖及白细胞介素-2(IL-2)mRNA表达水平的影响。结果表明,多糖单独加入到外周血淋巴细胞时,sTPS_(70c)和sCPPS_(50c)几乎所有浓度均可显著刺激T淋巴细胞的增殖(P0.05),而TPStp仅在浓度为3.125μg/mL时显著刺激T淋巴细胞增殖(P0.05);多糖与植物血凝素P(PHA-P)同时加入到外周血淋巴细胞时,sCPPS_(50c)在浓度为0.3911.563μg/mL时显著刺激T淋巴细胞增殖(P0.05);sTPS_(70c)和sCPPS_(50c)在浓度为1.563μg/mL时可提高T淋巴细胞IL-2 mRNA的表达水平(P0.05),其中sTPS_(70c)对IL-2 mRNA的促表达作用显著强于TPS_(tp)(P0.05),且在浓度为1.563μg/mL时的作用最强。结果提示,硫酸化修饰可以提高多糖的淋巴细胞增殖活性及显著增强IL-2 mRNA的表达,且以sTPS_(70c)的作用较强,这与取代度有一定的相关性。
[Abstract]:In order to study the immune enhancement mechanism of Tremella sulfated polysaccharide (sTPS_ (70c) and Codonopsis pilosula polysaccharide (sCPPS_ (50c), the effects of sTPS_ (70c) and sCPPS_ (50c) on the proliferation of chicken T lymphocytes and the expression of IL-2 (IL-2) mRNA were measured by thiazolyl blue (MTT) and real-time fluorescence quantitative PCR. The results showed that when polysaccharides were added to peripheral blood lymphocytes alone, almost all concentrations of sTPS_ (70c) and sCPPS_ (50c) could significantly stimulate the proliferation of T lymphocytes (P 0.05), while TPStp only stimulated the proliferation of T lymphocytes at the concentration of 3.125 渭 g / mL (P 0.05). When polysaccharide and plant hemagglutinin P (PHA-P) were added to peripheral blood lymphocytes at the concentration of 0.3911.563 渭 g / mL, sCPPS_ (50c) significantly stimulated the proliferation of T lymphocytes (P 0.05). When the concentration of sTPS_ (70c) and sCPPS_ (50c) was 1.563 渭 g / mL, the expression of IL-2 mRNA in T lymphocytes was increased (P 0.05), and the effect of sTPS_ (70c) on the expression of IL-2 mRNA was significantly stronger than that of TPS_ (tp) (P 0.05), and the effect was the strongest at the concentration of 1.563 渭 g / mL. The results suggest that sulfation modification can increase the proliferation activity of polysaccharide lymphocytes and significantly enhance the expression of IL-2 mRNA, and the effect of sTPS_ (70c) is strong, which is related to the degree of substitution.
