伊犁地区绵羊单增李氏杆菌病病原的分离鉴定及其快速检测技术的研究

【摘要】:李氏杆菌病(Listeriosis)是一种危害严重的食源性人畜共患传染病,单核细胞增多性李氏杆菌(Listeria monocytogenes,LM)简称单增李氏杆菌是导致其引发疾病的主要病原菌。现已证实该菌可感染40多种动物,主要可引起人和动物出现败血症、流产、脑膜炎等症状。近几十年,随着李氏杆菌病在世界范围内的不断暴发,已受到广泛关注。因此对各地区的绵羊进行单增李氏杆菌的分离鉴定,并建立单增李氏杆菌病的快速检测技术,对单增李氏杆菌病的预防和治疗具有重要的意义。根据国家标准(GB4789.30-2010)对伊犁部分地区(昭苏县、尼勒克县、霍城县、察布查尔县)羊场的绵羊、青贮饲料进行随机采样,通过培养特性、生化特性对分离株进行鉴定。利用伊犁地区绵羊体内分离鉴定的单增李氏杆菌作为平板凝集抗原,与标准阳性血清进行平板凝集试验,建立平板凝集检测方法,并通过特异性试验和符合性试验进行验证。结果从绵羊体内及青贮饲料中分离出14株单增李氏杆菌,表明抽查地区羊场的绵羊体内和青贮饲料中均存在一定程度的感染或污染。平板凝集试验确定平板凝集抗原最佳工作浓度为7×109CFU/m L,通过特异性试验证验该方法具有很好的特异性,符合性试验的总符合率为93.4%。研究结果表明伊犁地区的绵羊有发生单增李氏杆菌病的潜在威胁,建立的平板凝集方法可较为快速、准确的检测单增李氏杆菌病。以上研究结果可为伊犁地区绵羊的单增李氏杆菌病预防和监控提供科学的理论依据。
[Abstract]:Listeria monocytogenes (Listeriosis) is a serious food-borne zoonotic infectious disease. Listeria monocytogenes (Listeria monocytogenes,LM) is the main pathogen causing the disease. It has been confirmed that the bacteria can infect more than 40 kinds of animals, mainly causing septicemia, abortion, meningitis and other symptoms in humans and animals. In recent decades, with the continuous outbreak of listeriasis in the world, it has been widely concerned. Therefore, it is of great significance for the prevention and treatment of Listeria monocytogenes to isolate and identify Listeria monocytogenes in sheep and to establish a rapid detection technique for Listeria monocytogenes. According to the national standard (GB4789.30-2010), the sheep and silage of sheep farms in some areas of Yili (Zhaosu County, Nilek County, Huocheng County, Chabuchal County) were randomly sampled, and the isolates were identified by culture characteristics and biochemical characteristics. Using Listeria monocytogenes isolated and identified in Yili sheep as plate agglutination antigen, plate agglutination test was carried out with standard positive serum, and the plate agglutination detection method was established, which was verified by specific test and conformity test. Results 14 strains of Listeria monocytogenes were isolated from sheep and silage, which indicated that there was a certain degree of infection or pollution in sheep and silage in sheep farms. The optimum working concentration of plate agglutination antigen was determined to be 7 脳 109CFU/m / L by plate agglutination test. The method was proved to be very specific by specific test, and the total coincidence rate of conformance test was 93.4%. The results showed that sheep in Yili area had the potential threat of listeria monocytogenes, and the plate agglutination method could be used to detect listeria monocytogenes quickly and accurately. The above results can provide a scientific theoretical basis for the prevention and monitoring of listeria monocytogenes in sheep in Yili area.
