原儿茶酸在鸡组织中的残留消除规律

【摘要】:为研究原儿茶酸在鸡组织中残留消除规律,利用高效液相色谱法(HPLC)在260nm处检测原儿茶酸在鸡组织中残留情况,确定流动相体积比为乙腈∶水=9∶91,用H_3PO_4调pH至2.5。结果表明:试验测得鸡肝、肾、肌肉、皮肤和脂肪中检测限分别为13.5、25.0、12.0和16.0ng/g,定量限为45、85、40和50ng/g;平均加标回收率为63.30%~102.65%,变异系数为1.01%~11.43%。以临床推荐剂量20mg/kg(体重)口服给药,每天2次,连续5d,肾脏、肝脏中原儿茶酸的残留量最高,皮肤和脂肪、肌肉中残留量较低。休药4h后,各组织中原儿茶酸残留量均低于最高残留限量(肝脏1.52mg/g,肾脏3.05mg/g,肌肉0.51mg/g,皮肤和脂肪3.05mg/g)。试验结果表明,原儿茶酸在鸡组织中的残留消除较快,新药休药期的制定应根据具体情况进一步研究确定,目前初步建议休药期为0h。
