禽呼肠孤病毒S1133株在LMH细胞系中增殖特征的研究

【摘要】:为研究禽呼肠孤病毒(ARV)在鸡肝癌细胞(LMH)中的增殖特性,本研究将ARV S1133毒株感染LMH细胞,连续传代3次,观察细胞病变(CPE),通过RT-PCR鉴定病毒,并进一步研究病毒的增殖特性。结果显示,ARV S1133毒株能够在LMH细胞中很好地增殖,并出现典型的CPE。一步生长曲线结果显示,S1133感染LMH细胞后的前12 h为潜伏期,12 h~24 h为快速增长期,第24小时的效价达到最大,TCID50为10-9.2/0.1 m L。本研究为进一步研究ARV的致病机理和研制细胞灭活疫苗提供了良好的操作平台。
[Abstract]:In order to study the proliferation characteristics of avian reovirus (ARV) in chicken liver cancer cell line (LMH), LMH cells were infected with ARV S1133 strain for three consecutive generations. The cytopathic (CPE), was observed to identify the virus by RT-PCR, and the proliferation characteristics of the virus were further studied. The results showed that ARV S1133 strain could proliferate well in LMH cells and appeared typical CPE.. The results of one-step growth curve showed that the incubation period of LMH cells was 12 hours after S1133 infection, and the rapid growth period was 24 hours after S1133 infection. The titer of S1133 cells reached the maximum at 24 hours, and the titer of S1133 cells was 10 鈮,
