犬草酸钙结石形成机理的研究

草酸钙结晶在犬的尿液中检出率逐年升高,这几乎是世界各国都出现的问题。草酸或草酸根离子是形成含草酸钙结晶尿液的主要因素,草酸钙晶体继而可引起肾组织的严重损伤。坏死的。肾小管上皮细胞可作为晶体形成和聚集的核心,最终形成肾结石并且可导致肾衰。本研究通过给比格犬静脉注射KOX溶液,诱导草酸钙结晶尿和肾草酸盐沉积的模型,旨在探讨KOX对泌尿系统的损伤的作用。实验研究的内容如下:(1)KOX对犬血清和尿的生化指标影响,观测内容包括肝肾功能检测、尿沉渣检测和血清电解质浓度检测。(2)KOX对犬肾组织的形态学的影响,观测内容包括超声检查、病理组织学、扫描电镜(SEM)和透射电镜(TEM)观察。(3)KOX对犬肾骨桥蛋白(OPN)和肾小管蛋白(THP)表达的影响,通过免疫组化定位法和实时荧光PCR (qPCR)法检测OPN和THP的蛋白含量和基因表达量的变化。主要结果如下:(1)与对照组相比,处理组犬血清肌酐和尿素氮的含量在实验的0、1、3、5天逐渐增加,而尿液中肌酐和尿素氮的含量却逐渐减少。处理组犬的尿沉渣可检测到二水草酸钙(COD)和一水草酸钙(COM)晶体。(2)超声图像显示肾皮质和髓质组织的回声...
