东乡绿壳蛋鸡蛋壳绿色程度相关基因的鉴定

在鸡遗传研究和绿壳蛋鸡产业中,尤其是在鸡绿壳致因突变发现后,绿壳性状是一个有趣的研究对象。在中国,已经选育出东乡绿壳蛋鸡,且该群体中绿壳蛋的致因基因SLCO1B3的显性纯合子已经被固定。尽管被固定的绿壳蛋鸡都产绿壳蛋,但蛋壳的绿色程度却深浅不一。在本研究中我们对蛋壳颜色相关的色素胆绿素与原卟啉进行了基本分析,以此获得表型性状:胆绿素含量(QB),原卟啉含量(QP)及总色素含量(QT),然后我们测定了与血红素通路相关的三个潜在调控基因(SLCO1B3,H01,BLVRA)的mRNA表达水平,以评估这些候选基因与蛋壳颜色强度性状的相关性。我们的研究结果显示,这些基因与该性状没有显著的关联。在前期工作中,通过全基因组关联分析,我们鉴定了一些与蛋壳颜色深浅有关的18个SNP位点,为了验证全基因组关联分析的结果,我们在两个独立的中国东乡绿壳蛋鸡群体中对鉴定到的18个SNP进行了进一步的验证工作,这18个SNP均匀分布在GGA21与蛋壳色素显著相关的区域内。验证群体包括来自于用于GWAS研究群体产生的第二个世代,包含146个个体(N146),和另外一个来自江西东乡绿壳蛋鸡保种场的独立群体,包含28...
