RDS大鼠KLF2转录因子和KLF2 mRNA肺组织表达及甲状腺素水平变化研究

【摘要】:目的:1.检测不同时间点的RDS大白鼠和健康大白鼠的血清甲状腺素的变化规律。2.检测不同的时间点RDS大白鼠及健康大白鼠肺组织中KLF2蛋白和KLF2 mRNA的表达,并观察大鼠肺组织病理形态学改变。进而研究klf2转录因子在RDS大鼠肺组织的表达变化及甲状腺素水平的变化,探讨甲状腺素与klf2之间的相互作用,为临床用肺泡表面活性物质治疗不敏感的NRDS提供可能的理论依据。方法:SD雄性大鼠50只,随机分为4组,油酸组39只,再分为3小时、18小时和24小时组各13只,对照组11只不做任何处理。油酸组39只SD大鼠尾静脉注射0.17mg/Kg油酸建立RDS大鼠模型,分别于注射后3h、18h、24h用电化学发光法检测血清甲状腺素水平,同样用Western blot和RT-PCR方法检测RDS大白鼠以上时间段及非RDS大白鼠肺组织中KLF2蛋白和KLF2 mRNA的表达,并用组化免疫观察大鼠肺组织病理形态学改变。结果:1.油酸3h组、18h组中的血清FT4与正常对照组比较P0.01;油酸3h组中T4与正常对照组比较P0.01,有显著统计学意义;油酸18h组中T4数据与正常对照组比较P0.05,有统计学意义;2油酸3h组肺组织中的KLF2蛋白量与健康雄性SD大鼠肺组织中KLF2蛋白量数据对照比较,P0.05,有统计学意义;而18h组、24h无统计学意义,表明,在本组动物实验中,油酸所致RDS大鼠肺组织在3小时相比正常组KLF2蛋白量是减少的。3油酸24h组肺组织中的KLF2mRNA与健康雄性SD大鼠肺组织中KLF2m RNA数据对照比较,P0.05;有统计学意义;而3h组、18h组无统计学意义,表明,在本组动物实验中,油酸所致RDS大鼠肺组织在24小时相比正常组KLF2mRNA量有增多的趋势。结论:1.FT4、T4甲状腺激素在RDS大鼠3h、18h有明显下降趋势,呼吸窘迫综合症与血清甲状腺素水平有相关性。2.油酸所致RDS大鼠肺组织在3小时相比正常组KLF2蛋白量是减少的,呼吸窘迫综合症与KLF2蛋白有一定相关性。3.在RDS大鼠中血清FT4、T4是降低的,油酸3小时组肺组织KLF2蛋白量是减少的,因此FT4、T4与KLF2有一定的相关性。
[Abstract]:Objective: 1. To detect the changes of serum thyroxine in RDS rats and healthy rats at different time points. 2. The expression of KLF2 protein and KLF2 mRNA in lung tissues of RDS rats and healthy rats were detected at different time points, and the pathological changes of lung tissues were observed. Furthermore, the expression of klf2 transcription factors and the changes of thyroxine level in lung tissue of RDS rats were studied, and the interaction between thyroxine and klf2 was discussed, which provided a possible theoretical basis for the clinical treatment of insensitive NRDS with pulmonary surfactant. Methods: fifty male SD rats were randomly divided into 4 groups: oleic acid group (n = 39) and oleic acid group (n = 39). In the oleic acid group, 39 SD rats were injected with 0.17mg/Kg oleic acid into the tail vein to establish the model of RDS. The serum thyroxine levels were detected by electrochemiluminescence (ECL) at 3 h, 18 h and 24 h after injection, respectively. Western blot and RT-PCR methods were used to detect the expression of KLF2 protein and KLF2 mRNA in the lung tissues of RDS rats and non-RDS rats. The histopathological changes of lung tissues were observed by immunohistochemistry. Results: 1. The serum FT4 in 3 h group and 18 h group was significantly higher than that in normal control group (P 0.01), and that in 3 h group of oleic acid group was significantly higher than that in normal control group (P 0.01), and that in 3 h group of oleic acid group was significantly higher than that in control group. Compared with the normal control group, the content of KLF2 protein in the lung tissue of 3 h oleic acid group was significantly higher than that in the normal control group (P0.05), and the content of KLF2 protein in the lung tissue of the male SD rats was significantly higher than that in the control group (P0.05). In 18h group, there was no statistical significance at 24 h, which indicated that in this group of animal experiments, The content of KLF2 protein in lung tissue of RDS rats induced by oleic acid was lower than that of normal group at 3 hours. 3 KLF2mRNA in lung tissue of 24 h oleic acid group was compared with KLF2m RNA data in lung tissue of healthy male SD rats (P 0.05). However, there was no significant difference between the 3 h group and the 18 h group, which indicated that the lung tissue of oleic acid-induced RDS rats had a tendency to increase compared with the normal group at 24 hours. Conclusions: 1. FT4 T4 thyroid hormone decreased significantly in RDS rats at 3 h and 18 h, and respiratory distress syndrome was correlated with serum thyroxine level. 2. The content of KLF2 protein in lung tissue of RDS rats induced by oleic acid was decreased in 3 hours compared with that in normal group. Respiratory distress syndrome was associated with KLF2 protein. Serum FT4,T4 was decreased in RDS rats and KLF2 protein in lung tissue was decreased in 3 h oleic acid group. Therefore, there was a certain correlation between FT4,T4 and KLF2.
