人子宫颈癌肿瘤相关巨噬细胞浸润与T细胞亚型及临床病理特征的相关性分析

【摘要】:为了探讨肿瘤相关巨噬细胞(tumor associated macrophages,TAMs)在人子宫颈癌微环境内的分布与T细胞亚型和临床病理特征的相关性,本研究应用S-P免疫组化法检测TAMs在60例子宫颈癌组织、10例高级别鳞状上皮内病变组织及10例子宫颈组织的分布情况;间接免疫荧光双标法分析浸润的FOXP3~+Treg细胞数量、CD8~+T细胞数量及其粒酶B分泌情况。统计学分析TAMs数量与癌间质浸润的FOXP3~+Treg、CD8~+T数量和临床病理特征的相关性。结果显示TAMs在子宫颈癌微环境的数量均显著多于高级别鳞状上皮内病变和子宫颈组织(p0.01,p0.01),同时高级别鳞状上皮内病变组织阳性细胞数也明显多于子宫颈组织(p0.01);TAMs数量与宫颈癌间质内浸润的FOXP3~+T细胞数量呈正相关性(p0.05),且与淋巴结转移和癌栓相关(p0.05,p0.05),但与CD8~+T细胞浸润数量无明显相关性(p0.05)。本实验发现TAMs在宫颈癌微环境内大量分布,且与肿瘤局部免疫抑制及淋巴结转移和癌栓有关。
[Abstract]:In order to study tumor associated macrophage tumor associated macrophages. The distribution of Tams in the microenvironment of human cervical cancer was correlated with T cell subtypes and clinicopathological features. S-P immunohistochemical method was used to detect TAMs in 60 cases of cervical carcinoma. The distribution of 10 cases of high grade squamous intraepithelial lesions and 10 cases of cervical tissues. Indirect immunofluorescence double labeling method was used to analyze the number of FOXP3- Treg cells. The number of CD8 ~ T cells and the secretion of granulocyte B. the number of TAMs and FOXP3- Treg of interstitial invasion were analyzed statistically. The correlation between the number of CD8T and clinicopathological features. The results showed that the number of TAMs in the microenvironment of cervical cancer was significantly higher than that in high-grade squamous intraepithelial lesions and cervical tissue (p0.01). At the same time, the number of positive cells in high grade squamous intraepithelial lesions was significantly higher than that in cervical tissues. The number of TAMs was positively correlated with the number of FOXP3T cells infiltrated in the stroma of cervical carcinoma, and was correlated with lymph node metastasis and tumor thrombus. However, there was no significant correlation between the number of CD8T cells and the number of CD8T cells. In this study, we found that TAMs was widely distributed in the microenvironment of cervical cancer. And related to tumor local immunosuppression, lymph node metastasis and tumor thrombus.
