【摘要】：目的:通过检测不同胚胎质量的体外受精(in vitro fertilization,IVF)患者精子mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375的表达情况,探讨精子来源的mi RNAs与胚胎质量的相关性。方法:选择2012年9月至12月在中南大学湘雅医院生殖医学中心行IVF治疗原发不育的男性患者44例,收集IVF取卵当日剩余新鲜精液标本,采用实时荧光定量PCR测定精子mi RNAs(mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375)的表达水平。观察胚胎情况后分组,第3天胚胎评分的平均值8分为实验组,≥8分为对照组。比较实验组和对照组患者一般情况及实验室资料,分析精子mi RNAs表达水平与胚胎质量的相关性。结果:实验组精子mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375表达水平低于对照组(P0.01)。实验组与对照组的获卵数、减数分裂II期(metaphase II,MII)卵子数、双原核(dual pronuclear,2PN)受精数、卵裂数、受精率差异无统计学意义(P0.05);实验组卵裂率低于对照组(P0.05)。精子mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375表达水平与第2,3天胚胎碎片率呈负相关,与第3天胚胎卵裂球数呈正相关,与胚胎评分呈正相关。结论:精子mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375的表达水平上升可能影响卵裂期胚胎质量,对胚胎发育可能起一定的积极作用。
