HPVL1蛋白与IL-2在宫颈病变中的表达及其相关性

【摘要】：目的检测HPVL1蛋白和IL-2在宫颈病变中的表达,探讨二者与宫颈病变的的关系和临床意义,为癌前病变及宫颈癌的诊断和治疗提供实验依据。 方法选取2011年4月至2011年9月在山西省肿瘤医院就诊的宫颈液基细胞学检查诊断≥未明确诊断意义的不典型鳞状细胞(ASCUS),且检测HPVDNA均为阳性,同时有组织病理学诊断(作为金标准)患者的宫颈组织石蜡包埋标本,采用免疫组织化学法结合应用计算机图像分析方法对检测结果进行图像采集,并对实验数据进行统计学分析,观察HPVL1蛋白和IL-2在宫颈上皮内瘤病变及宫颈癌中的表达情况,从而研究HPVL1蛋白和IL-2与宫颈上皮内瘤病变及宫颈癌之间的关系。结果免疫组织化学方法显示：HPVL1蛋白的表达定位于细胞核,主要表达于宫颈组织的上皮层。159例标本中,HPVL1阳性表达为47.80%(76例)。慢性宫颈炎、CINI、 CINⅡ、CINIII及SCC的阳性表达分别为62.50%(30/48)、83.33%(35/42)、37.03%(10/27)、3.3%(1/30)、0%(0/12)。除了CINIII与SCC比较,差异无统计学意义(P0.05)外,其他不同病变间的比较,差异有统计学意义(P均0.05)。IL-2表达定位于细胞膜和/或细胞质,主要表达于宫颈组织的上皮层。IL-2阳性表达为64.78%(103例)。慢性宫颈炎、CINⅠ、CINⅡ、CINIII及SCC的阳性表达分别为83.33%(40/48)、89.30%(37/42)、62.97%(17/27)、3.3%(9/30)及0%(0/12),差异均有统计学意义(P0.05)。结论本实验研究结果显示,在宫颈上皮内瘤变组织中,HPVL1蛋白与IL-2的表达具有相关性(rs=0.165, p0.05)。HPVL1-/IL-2-的表达44例,其表达随着宫颈病变的程度的增加而呈上升趋势,HPVL1+/IL-2+的表达64例,其表达随着宫颈病变的程度的增加而呈下降趋势。
[Abstract]:Objective to detect the expression of HPVL1 protein and IL-2 in cervical lesions and to explore the relationship between them and cervical lesions and to provide experimental evidence for the diagnosis and treatment of precancerous lesions and cervical cancer. Methods from April 2011 to September 2011, cervical liquid-based cytology was performed in Shanxi Provincial Cancer Hospital. Atypical squamous cells with no definite diagnostic significance were selected, and HPVDNA was positive. At the same time, paraffin embedded specimens of cervical tissues of patients with histopathological diagnosis (as gold standard) were collected by immunohistochemical method and computer image analysis. The expression of HPVL1 protein and IL-2 in cervical intraepithelial neoplasia and cervical carcinoma were analyzed statistically. The relationship between HPVL1 protein and IL-2 and cervical intraepithelial neoplasia and cervical carcinoma was studied. Results the expression of HPVL1 L1 protein was localized in the nucleus by immunohistochemical method. The positive expression of HPVL1 was 47.80 in 76 cases of cervical epithelium.
