胎盘中激活素受体的表达及与胎儿生长受限的关系

【摘要】:目的: 检测激活素(Activin)受体在胎儿生长受限胎盘及正常足月胎儿胎盘中的定位及表达水平,研究其与胎儿生长受限的关系,探讨激活素受体在胎儿生长受限发病机制中的作用。 方法: 各收集15例足月胎儿生长受限和正常足月妊娠孕妇分娩后的胎盘组织。HE染色法分析各组胎盘组织;免疫组织化学法分析各组胎盘组织中激活素受体的表达部位,并进行半定量分析;Real-Time PCR法测定各组胎盘组织中激活素受体基因mRNA的表达。 结果: 1、HE染色结果:对照组胎盘组织绒毛发育良好,,绒毛血管扩张明显,间质细胞少,绒毛滋养细胞以合体滋养细胞为主;FGR组胎盘组织绒毛发育欠佳,绒毛血管扩张不良,绒毛内间质增生,钙化、坏死增多。 2、免疫组化染色结果:两组胎盘组织中激活素受体均主要表达于合体滋养细胞及细胞滋养细胞胞浆中。FGR组胎盘组织中激活素受体ⅠA、ⅠB染色阳性程度与对照组相比,差异无统计学意义(P>0.05);激活素受体ⅡA、ⅡB染色阳性程度均显著高于对照组,差异有统计学意义(P<0.05)。 3、Real-Time PCR结果:FGR组胎盘组织中激活素受体ⅠA、Ⅰ B基因表达与对照组相比,差异无统计学意义(P>0.05);激活素受体Ⅱ A、 ⅡB基因表达均较对照组升高,差异有统计学意义(P<0.05)。 结论: 1、胎儿生长受限患者胎盘组织中激活素受体ⅠA、Ⅰ B表达水平与正常妊娠相比无差异,激活素受体ⅡA、Ⅱ B表达水平较正常妊娠明显升高。 2、妊娠晚期胎盘组织中激活素受体ⅡA、 ⅡB表达水平的上升可能是导致胎儿生长受限发生的一个重要因素。 3、激活素及其受体共同参与调节胎儿宫内生长发育,激活素受体的紊乱可能在胎儿生长受限的发病机制中发挥重要的作用。
[Abstract]:Objective:. To detect the localization and expression of Activinin receptor in placenta of fetal growth restriction and normal term fetal placenta, to study the relationship between activin receptor and fetal growth restriction, and to explore the role of activin receptor in the pathogenesis of fetal growth restriction. Methods:. The placental tissues of 15 pregnant women with full-term fetal growth restriction and normal term pregnancy were collected. The placenta tissues were analyzed by HE staining, and the expression sites of activin receptors in placenta tissues were analyzed by immunohistochemical method. The expression of activin receptor gene (mRNA) in placenta was determined by semi-quantitative analysis with Real-Time PCR. Results:. 1the results of HE staining showed that the placental villi of the control group developed well, the villus vasodilated obviously, the interstitial cells were few, the villus trophoblast was mainly composed of syncytiotrophoblast and the villi of the FGR group were not well developed and the villi were dilated poorly. Interstitial hyperplasia, calcification and necrosis in villi. 2. Immunohistochemical staining: Activin receptors were mainly expressed in the cytoplasm of syncytiotrophoblast and cytotrophoblast.
