人类黑色素瘤相关抗原A3在宫颈癌中的表达及其临床意义

【摘要】:目的 检测宫颈正常组织、宫颈上皮内瘤变3级(cervical intraepithelial neoplasias,CIN3)及宫颈癌组织中人类黑色素瘤相关抗原A3(MAGE-A3)的表达,分析该基因在各组织中的变化与临床意义,并探讨该基因在宫颈癌发生发展及转移过程中的作用及分子机制。方法 取2015年1月至2016年4月河北大学附属医院采集的72例宫颈组织,病理切片诊断患者宫颈组织类型,判断分化程度,应用实时荧光定量PCR(q-PCR)和Western印迹法检测不同宫颈组织中MAGE-A3基因及其蛋白的表达水平。结果 q-PCR结果提示,宫颈癌组MAGE-A3基因表达量明显高于正常宫颈组及CIN3组,三组间差异均有统计学意义(P0.05);Western印迹检测MAGE-A3蛋白在正常宫颈组织中的阳性表达率为20%,在CIN3中为36.7%,在宫颈癌(Ⅰ期、ⅡA期)组织中为61.1%,三组间差异有统计学意义(P=0.002);其中ⅡA期MAGE-A3蛋白表达率明显高于Ⅰ期的宫颈癌组织(P=0.038)。MAGE-A3蛋白在宫颈癌组织中的表达与病理类型、肿瘤大小、绝经状态均无明显相关性(P0.05)。结论 MAGE-A3的表达与宫颈癌的分化级别、临床分期及有无淋巴结转移情况呈正相关趋势,该基因可能在宫颈癌的发生发展及转移过程中起着重要的作用。
[Abstract]:Objective to detect the expression of human melanoma associated antigen A3 (MAGE-A3) in normal cervical tissues, cervical intraepithelial neoplasia grade 3 (grade 3) and cervical carcinoma tissues, and to analyze the changes and clinical significance of this gene in all tissues. To explore the role and molecular mechanism of the gene in the development and metastasis of cervical cancer, 72 cases of cervical tissues collected from the affiliated Hospital of Hebei University from January 2015 to April 2016 were collected, and the pathological sections were used to diagnose the cervical tissue types of the patients. The expression of MAGE-A3 gene and its protein in different cervical tissues were detected by real-time fluorescence quantitative PCR q-PCR and Western blotting. Results the results of q-PCR showed that the expression of MAGE-A3 gene in cervical cancer group was significantly higher than that in normal cervix group and CIN3 group.
