葡萄胎病理相关新基因F10与绒癌细胞系JEG-3成瘤性的关系

【摘要】:目的探讨葡萄胎病理相关新基因F10与绒癌细胞系JEG-3裸鼠皮下成瘤的关系。方法通过细胞转染技术及RNA干扰技术,分别建立和筛选出F10基因稳定过表达及沉默的绒癌细胞系JEG-3。取SPF级裸鼠(4~5周龄)30只,随机均分为JEG-3F10过表达组、JEG-3 F10沉默组、JEG-3未处理组(n=10),分别接种F10基因过表达的JEG-3细胞、F10基因沉默的JEG-3细胞和未处理的JEG-3细胞株5×107个于颈背部皮下。接种后每3~4 d称量裸鼠质量观察,记录肿瘤发生时间,观察皮下肿瘤的生长情况。绘制在体肿瘤生长曲线,计算各组裸鼠的成瘤率。结果 3组的成瘤率均为100%(10/10)。JEG-3 F10过表达组、JEG-3F10沉默组、JEG-3未处理组的成瘤时间分别为6.2±0.78、7±2.49、6.3±0.67 d,组间比较无统计学差异(F=0.781,P=0.468)。JEG-3 F10过表达组肿瘤的在体生长速度较JEG-3 F10沉默组、JEG-3未处理组增快,差异有统计学意义(P0.05),JEG-3 F10未处理组肿瘤的在体生长速度较JEG-3 F10沉默组快(P0.05)。细胞接种5周后处死小鼠,取瘤组织称重,JEG-3 F10过表达组、JEG-3 F10沉默组、JEG-3未处理组肿瘤重量分别为571.1±221.10 mg、136.2±66.25 mg、354.5±116.23 mg,组间比较存在统计学差异(F=21.199,P=0.000)。结论 F10基因与绒癌细胞系JEG-3的增殖调节有关,可增强JEG-3细胞系在裸鼠体内的致瘤性。
[Abstract]:Objective to investigate the relationship between a novel histopathological gene F10 and subcutaneous tumorigenesis of choriocarcinoma cell line JEG-3 in nude mice. Methods A choriocarcinoma cell line JEG-3 with stable overexpression and silencing of F10 gene was established and screened by cell transfection and RNA interference. Thirty SPF grade nude mice were randomly divided into JEG-3F10 overexpression group (JEG-3 F10 silencing group) and JEG-3 untreated group (JEG-3 untreated group). JEG-3 cells silenced by F10 gene over-expressed JEG-3 cells and 5 脳 107 untreated JEG-3 cell lines were inoculated subcutaneously on the neck and back respectively. The quality of nude mice was measured every 3 ~ 4 days after inoculation, the occurrence time of tumor was recorded and the growth of subcutaneous tumor was observed. The tumor growth curve in vivo was drawn and the tumorigenic rate of each group was calculated. Results the tumorigenesis rate of the three groups was 6.2 卤0.787 卤2.49 卤0.67 days in the 100%(10/10).JEG-3 F10 overexpression group and JEG-3F10 silencing group, respectively. There was no significant difference between the three groups. The growth rate of the tumor in the overexpression group was faster than that in the untreated group of JEG-3 F10 silencing group. The in vivo growth rate of JEG-3 F10 untreated group was faster than that of JEG-3 F10 silencing group (P 0.05). After 5 weeks of cell inoculation, the mice were killed. The tumor weight of the untreated JEG-3 F10 group was 571.1 卤221.10 mg / g, respectively, and the tumor weight was 571.1 卤221.10 mg / kg, 354.5 卤116.23 mg / g respectively. There was a significant difference between the two groups in F21.199P0.000mg. Conclusion F10 gene is related to the proliferation regulation of choriocarcinoma cell line JEG-3 and can enhance the tumorigenicity of JEG-3 cell line in nude mice.
