β-catenin在卵巢子宫内膜异位症在位及异位内膜间质细胞的表达及意义

【摘要】:目的: 体外分离培养卵巢子宫内膜异位症患者在位内膜及异位内膜间质细胞,并探讨-catenin在卵巢子宫内膜异位症在位及异位子宫内膜间质细胞的表达及其意义。 方法: 使用胶原酶消化联合二次筛网过滤法体外分离、纯化及培养卵巢子宫内膜异位症患者在位、异位和对照组子宫内膜间质细胞,分别应用逆转录聚合酶链反应(RT-PCR)、Wester-blot方法检测上述各组细胞中-catenin的mRNA和蛋白表达情况。 结果: 分离得到的内膜间质细胞纯度达95%。OEms在位内膜、异位内膜和对照组中-catenin mRNA的IOD值分别为0.672±0.07,0.351±0.029和0.184±0.020,各组间均有统计学差异(F=27.68,P<0.001)。OEms在位内膜、异位内膜和对照组中-catenin蛋白的IOD值分别为0.803±0.074,,0.388±0.044和0.186±0.026,各组间均有统计学差异(F=36.83,P<0.001)。-catenin mRNA和蛋白的表达均在OEms在位内膜最高,异位内膜次之。 结论: 成功体外分离培养原代子宫内膜间质细胞,鉴定其纯度达95%,可以为EMs的研究提供理想模型。OEMs在位、异位内膜及正常对照组中均有β-cateninmRNA和蛋白的表达;β-catenin在在位内膜和异位内膜的表达明显高于正常对照组,其中在位内膜表达最高。-catenin分子可能在EMs在位内膜生物学改变及异位内膜完成“三部曲”(即黏附、侵袭和血管形成)中起着重要作用,与EMs的发生发展密切相关。
[Abstract]:Objective: The eutopic endometrium and ectopic endometrial stromal cells were isolated from patients with ovarian endometriosis in vitro. The expression and significance of -catenin in eutopic and ectopic endometrial stromal cells of patients with ovarian endometriosis were investigated. Methods: The eutopic, ectopic and control endometrial stromal cells of patients with ovarian endometriosis were purified and cultured by collagenase digestion combined with secondary screen filtration in vitro. The mRNA and protein expression of -catenin were detected by reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot. Results: The purity of the isolated endometrial stromal cells was as high as that of 95%.OEms eutopic endometrium. The IOD values of -catenin mRNA in ectopic endometrium and control group were 0.672 卤0.07 卤0.351 卤0.029 and 0.184 卤0.020, respectively. There was significant difference between the two groups (P < 0.001).OEms). The IOD values of -catenin protein in ectopic endometrium and control group were 0.803 卤0.074 卤0.388 卤0.044 and 0.186 卤0.026, respectively. There was a significant difference in the expression of -catenin mRNA and protein between ectopic endometrium and ectopic endometrium. The expression of -catenin mRNA and protein in ectopic endometrium was the highest in OEms eutopic endometrium, followed by ectopic endometrium. Conclusion: The primary endometrial stromal cells were successfully isolated and cultured in vitro, and their purity was 95%, which could provide an ideal model for the study of EMs. The expression of 尾 -catenin mRNA and protein in ectopic endometrium and normal control group was significantly higher than that in eutopic endometrium and ectopic endometrium, and the expression of 尾 -catenin in eutopic endometrium and ectopic endometrium was significantly higher than that in normal control group. The highest expression of .-catenin in eutopic endometrium may play an important role in the biological changes of eutopic endometrium and the completion of "trilogy" (adhesion, invasion and angiogenesis) in eutopic endometrium of EMs, which is closely related to the occurrence and development of EMs.
