MicroRNA-182在宫颈癌中的表达及对肿瘤细胞增殖、侵袭及迁移的影响

【摘要】:目的 探讨Micro RNA(mi R-182)在宫颈癌组织、宫颈上皮内瘤变组织(CIN)和正常宫颈组织中的表达,及其对宫颈癌细胞增殖、侵袭和迁移能力的影响。方法 应用实时聚合酶链反应(real-time PCR)技术方法检测78例宫颈癌组织、30例CIN组织及20例正常宫颈组织中mi R-182的表达,并分析其与宫颈癌临床病理特征之间的关系。在宫颈癌细胞株Ca Ski中,应用细胞转染技术上调mi R-182的表达,应用Cell Counting Kit-8(CCK-8)试剂盒检测细胞增殖能力的变化,Transwell实验检测细胞侵袭迁移能力的变化。结果 Realtime PCR结果显示mi R-182在宫颈癌组织、宫颈上皮内瘤变(CIN)组织与正常宫颈组织比较,差异有统计学意义P0.05㖞;两两比较,mi R-182在宫颈癌组织和CIN组织中的表达较正常宫颈组织降低P0.05㖞,mi R-182在宫颈癌组织与CIN组织比较,差异无统计学意义(P0.05),宫颈癌组织中mi R-182的表达与分化程度、淋巴转移密切相关,而与年龄、肿瘤直径、浸润深度及临床分期等无关(P0.05)。在人宫颈癌上皮细胞(Ca Ski)中,上调mi R-182表达能够抑制肿瘤细胞的侵袭和迁移P0.05㖞,但不改变细胞的增殖能力。结论 mi R-182在宫颈癌组织中低表达,mi R-182的表达水平与分化程度和淋巴结转移有关,上调mi R-182的表达能够抑制宫颈癌Ca Ski细胞的迁移及侵袭。
[Abstract]:Objective to investigate the expression of Micro RNA (MI R-182) in cervical cancer tissue, cervical intraepithelial neoplasia (CIN) and normal cervical tissue, and its effect on the proliferation, invasion and migration of cervical cancer cells. Methods the real-time polymerase chain reaction (real-time PCR) technique was used to detect 78 cervical cancer tissues, 30 CIN tissues and 20 normal cases. The expression of MI R-182 in cervical tissue and its relationship with the clinicopathological features of cervical cancer. In Ca Ski of cervical cancer cell line, the expression of MI R-182 was up-regulated by cell transfection technique, the changes of cell proliferation ability were detected by Cell Counting Kit-8 (CCK-8) kit, and the changes of cell invasion and migration ability were detected by Transwell experiment. Results Realtime PCR results showed that MI R-182 was compared with normal cervical tissue in cervical cancer tissue, cervical intraepithelial neoplasia (CIN) tissue and normal cervical tissue, the difference was statistically significant? P0.05? (22) the expression of MI R-182 in cervical cancer tissue and CIN tissue was lower than normal cervical tissue? P0.05? Mi R-182 in cervical cancer tissue compared with the tissue of cervical cancer. There is no statistical significance (P0.05). The expression of MI R-182 in cervical cancer tissues is closely related to the degree of differentiation and lymphatic metastasis, but not related to age, tumor diameter, depth of invasion and clinical staging (P0.05). In human cervical cancer epithelial cells (Ca Ski), up regulation of MI R-182 can inhibit the invasion and migration of tumor cells? P0.05? Conclusion the expression of MI R-182 in cervical cancer tissues is low, and the expression level of MI R-182 is related to the degree of differentiation and lymph node metastasis. The expression of MI R-182 can inhibit the migration and invasion of Ca Ski cells in cervical cancer.
