缺氧诱导因子-1α与缺氧诱导因子-2α蛋白和mRNA在宫颈癌中的表达及其临床意义

【摘要】:目的探讨缺氧诱导因子(HIF)-1α和HIF-2α在宫颈癌中的表达及其与患者临床病理特征的相关性。方法用免疫组化法、蛋白印迹(Western blot)和实时定量聚合酶链式反应(RT-PCR)技术检测30例宫颈癌患者(试验组)癌组织及30例宫颈良性病变患者(对照组)组织中HIF-1α和HIF-2α蛋白及mRNA的表达,并分析其与宫颈癌临床病理特征的相关性。结果 HIF-1αmRNA,HIF-2αmRNA在试验组中相对表达量为对照组的(2.06±0.68),(1.62±0.43),差异均有统计学意义(均P0.01)。免疫组化结果显示,试验组中HIF-1α和HIF-2α的阳性表达率分别为63.33%和70.00%,对照组分别为16.67%和13.33%,差异均有统计学意义(均P0.01)。Western blot提示,试验组HIF-1α和HIF-2α蛋白表达量分别为(0.71±0.01)和(0.71±0.03),对照组分别为(0.53±0.03)和(0.56±0.05),差异均有统计学意义(均P0.05)。HIF-1α表达与淋巴结转移及放疗前贫血相关(均P0.05),HIF-2α表达与放疗前贫血相关(P0.05)。结论试验组中,HIF-1α、HIF-2α在mRNA及蛋白质水平均出现明显升高,在宫颈癌的发展过程中可能起促癌作用;且两者的表达呈同向变化,提示他们可能通过不同机制起着相同或相仿的作用。
[Abstract]:Objective to investigate the expression of HIF-1 伪 and HIF-2 伪 in cervical carcinoma and their correlation with clinicopathological features. Methods the expressions of HIF-1 伪 and HIF-2 伪 and mRNA were detected by immunohistochemistry, Western blot and real-time quantitative polymerase chain reaction (RT-PCR) in 30 patients with cervical cancer (trial group) and 30 patients with benign cervical lesions (control group). The relationship between the clinicopathological features of cervical cancer and its clinical features was analyzed. Results the relative expression of HIF-1 伪 mRNA-HIF-2 伪 mRNA in the experimental group was 2.06 卤0.68, 1.62 卤0.43, and the difference was statistically significant (all P 0.01). The results of immunohistochemistry showed that the positive rates of HIF-1 伪 and HIF-2 伪 were 63.33% and 70.005% in the experimental group and 16.67% and 13.33% in the control group, respectively. The expressions of HIF-1 伪 and HIF-2 伪 in the experimental group were 0.71 卤0.01 and 0.71 卤0.03, respectively, and those in the control group were 0.53 卤0.03 and 0.56 卤0.05, respectively. The difference was statistically significant (all the expressions of P0.05).HIF-1 伪 were correlated with lymph node metastasis and anemia before radiotherapy (all P0.05HIF-2 伪 expression was associated with anemia before radiotherapy). Conclusion in the experimental group, the level of HIF-1 伪 and HIF-2 伪 increased significantly in mRNA and protein levels, which may play a promoting role in the development of cervical cancer, and the expression of HIF-1 伪 and HIF-2 伪 may play the same or similar role in the development of cervical cancer, suggesting that they may play the same or similar role through different mechanisms.
