NDRG4在宫颈癌细胞中的表达及其对增殖的影响

【摘要】:目的:研究N-MYC下游调控基因4在宫颈癌细胞中的表达及其对宫颈癌细胞增殖、凋亡及周期的影响。方法:通过荧光实时定量聚合酶链反应和免疫印迹检测HeLa细胞中NDRG4的表达,构建NDRG4稳转细胞系HeLa-NDRG4。CCK-8法检测NDRG4对Hela细胞增殖的影响,流式细胞术检测NDRG4对Hela细胞凋亡及周期的影响。结果:宫颈癌细胞中NDRG4 m RNA和蛋白表达水平均较人正常宫颈内皮细胞显著降低,HeLa-NDRG4细胞的增殖能力显著低于HeLa-MOCK细胞,而凋亡能力显著高于HeLa-MOCK细胞。与HeLa-MOCK细胞相比,HeLa-NDRG4细胞周期被阻滞于G1期。HeLa-NDRG4细胞中p-Akt的蛋白水平较HeLa-MOCK细胞显著降低,而PTEN的蛋白水平较HeLa-MOCK细胞显著升高。结论 :NDRG4在宫颈癌中作为抑癌基因发挥作用。
[Abstract]:Aim: to investigate the expression of N-MYC downstream regulatory gene 4 in cervical cancer cells and its effects on cell proliferation, apoptosis and cell cycle. Methods: the expression of NDRG4 in HeLa cells was detected by fluorescence real-time quantitative polymerase chain reaction and Western blotting. The effect of NDRG4 on the proliferation of Hela cells was detected by HeLa-NDRG4.CCK-8 method, and the effect of NDRG4 on apoptosis and cell cycle of Hela cells was detected by flow cytometry. Results: the expression levels of NDRG4 m RNA and protein in cervical cancer cells were significantly lower than those in normal cervical endothelial cells. The proliferative ability of HeLa-NDRG4 cells was significantly lower than that of HeLa-MOCK cells, and the apoptotic ability was significantly higher than that of HeLa-MOCK cells. Compared with HeLa-MOCK cells, the cell cycle of HeLa-NDRG4 cells was blocked in G1 phase. The protein level of p-Akt in He-La-NDRG4 cells was significantly lower than that of HeLa-MOCK cells, while the protein level of PTEN was significantly higher than that of HeLa-MOCK cells. Conclusion the expression of the tumor suppressor gene in cervix carcinoma may be affected by the expression of 1% NDRG 4.
